三种大鼠肌肉组织蛋白提取方法的比较研究

The extraction methods of tissue protein were established and optimized.The tis-sue proteins of rats were extracted by TCA/acetone, improved TCA/acetone and tissue pro-tein extraction kit.Then the optical density of extracted proteins was detected and the protein was further determined using sodium dodecyl sulfate polyacrylamide gel electrophoresis.UV spectrum analysis showed that the protein content is lower at 562 nm using TCA/acetone method but higher using improved the TCA/acetone and tissue protein extraction kit method. Results of sodium dodecyl sulfate polyacrylamide gel electrophoresis confirmed that the pro-tein fragments of TCA/acetone method was the weakest among them.The protein fragments of the others were clear, and there were no degradation or trailing phenomena.%建立并优化组织蛋白的提取方法.以大鼠肌肉组织为实验材料,分别采用三氯乙酸( TCA)/丙酮沉淀法、改良的TCA /丙酮沉淀法和试剂盒法对其组织蛋白进行提取,对三种方法提取的蛋白进行光密度、聚丙烯酰胺凝胶电泳检测.紫外光谱检测表明TCA/丙酮沉淀法在波长562 nm蛋白含量比较低,而改良的TCA /丙酮沉淀法和试剂盒法的蛋白含量比较高,聚丙烯酰胺凝胶电泳检测证实:常规的TCA/丙酮沉淀法提取的蛋白条带模糊,其他两种方法的提取的蛋白条带很清晰,蛋白没有降解,无拖尾现象.
