清热活血调气方对坏死性小肠结肠炎模型新生大鼠肠损伤的影响

     

摘要

Objective To explore the protective effect of Qingre Huoxue Tiaoqi Recipe (QHTR) on intestinal injury in neonatal rats necrotizing enterocolitis ( NEC) model. Methods Thirty-six three-day-old neonatal Sprague-Dawley (SD) rats were randomly divided into three groups: normal control group, NEC model group and treatment group, 12 in each group. NEC rats model was established by feeding with rat-milk substitutes, and by exposing to 100% nitrogen gas for 90s and cold stress 4℃ (for 10 min) twice daily for 3 days. The rats in the treatment group received Chinese herbal preparation QHTR mixed into the milk substitutes (30mL/kg each time, 3 times per day) , and were given hypoxia and cold stress. Rats in the normal control group were given cold stress and fed with milk of their mother. On the 4th day, all the subjects were sacrificed and intestinal tissues were isolated. The histological changes of proximal intestines of ileocecal junction were scored by double-blind way after hematoxylin-eosin (HE) staining, and the rats with mean histological scores over 2 were defined as NEC. Results Compared with the normal control group, the histological scores of the model group were significantly increased, and body weight was decreased (P < 0.01). Compared with the model group, the pathological scores and incidence of NEC were decreased, and body weight was increased in the treatment group (P < 0. 05 or P < 0. 01). Conclusion QHTR has protective effect against NEC in neonatal rats model.%[目的]探讨清热活血调气方对坏死性小肠结肠炎( necrotizing enterocolitis,NEC)模型新生大鼠肠损伤的保护作用.[方法]选用SPF级3日龄Sprague-Dawley新生大鼠36只,随机分为正常对照组、NEC模型组及治疗组,每组12只.NEC模型组采用鼠代乳品人工喂养、体积分数100%氮气缺氧90s、4℃冷刺激10 min方法,每天2次,连续3d,复制新生大鼠NEC模型;治疗组采用代乳品+清热活血调气方(30 mL·kg-1·次-1,3次/d)人工喂养,同时予缺氧冷刺激;正常对照组置于母鼠身旁喂养,不进行任何干预.各组于造模后第4天处死大鼠取出肠组织,取近回盲部肠组织做病理切片,观察组织学变化并做组织损伤评分,组织学评分≥2为NEC.[结果]与正常对照组比较,模型组肠损伤组织病理评分显著升高,体质量显著下降(P<0.01);与模型组比较,治疗组肠损伤组织病理评分与发病率显著降低,体质量显著升高(P<0.05或P<0.01).[结论]清热活血调气方对NEC模型新生大鼠肠损伤具有一定保护作用.
