当归芍药散及其有效部位对认知障碍模型小鼠学习记忆功能的影响

         

摘要

目的 研究当归芍药散及其有效部位对D-半乳糖结合AlCl3致认知障碍模型小鼠学习记忆功能的影响.方法 取2~3个月龄的小鼠,以D-半乳糖联合AlCl3造认知障碍动物模型,用不同浓度的当归芍药散及其有效部位对建模成功的小鼠进行腹腔注射,用Y-型迷宫进行学习记忆能力测试,检测后测定小鼠血清中SOD、MDA、GSH-Px和脑内脂褐素的含量,脑组织行HE染色,观察其病理变化.结果 当归芍药散及其有效部位组的小鼠学习能力和记忆能力均显著优于模型组(P<0.01);与模型组比较,当归芍药散及其有效部位组明显升高损伤小鼠血中SOD活性(P<0.01),降低损伤小鼠血中MDA(P<0.01),明显升高损伤的小鼠血中GSH-Px活性(P<0.01),明显降低小鼠脑中脂褐素的含量(P<0.01);当归芍药散及其有效部位能够改善模型鼠脑组织病理损伤情况.结论 当归芍药散及其有效部位能够有效改善该认知障碍模型小鼠的学习和记忆能力,并且具有较为显著的抗氧化能力,对该模型小鼠脑组织损伤具有一定的保护作用.%Objective To study the effect of Danggui Shaoyao San ( DSS ) and its active compositions on learning and memory behaviors in cognitive impairment mice induced by D-galactose and AlCl3 . Methods The model was established by administration of D-galactose and AlCl3 in mice. DSS and its active compositions were used at different doses. Y-maze trial was performed to evaluate their learning and memory behaviors. The contents of SOD, MDA, GSH-Px in serum and lipofuscin in brain were detected, and brain tissues were stained with HE and observed. Results Compared with the model mice, DSS and its active compositions improved learning and memory behaviors significantly ( P<0. 01 ) . Meantime, DSS and its active compositions increased the activities of serum SOD and GSH-Px, but decreased the levels of serum MDA and brain lipofuscin ( P<0. 01 ) . DSS and its active compositions also improved the pathological damage of brain tissue in model mice. Conclusion DSS and its active compositions effectively improve the learning and memory behaviors in cognitive impairment mice, which may be related to its antioxidant abilities.
