白藜芦醇对大鼠脊髓损伤的保护作用

     

摘要

AIM: To study the protective effects of Resveratrol (Res) on spinal cord injury in rats. METHODS: Twenty-four SD rats were randomly divided into 3 groups: Sham-operation group( group A ), spinal cord injury group ( group B ) and Res group (group C). Animal models of acute spinal cord were induced with Allen's method. In RES group, Res of 100 mg/kg was injected through abdomen immediately after modeling. The locomotor function of the hind limbs and the histopathological as well as superoxide dismutase(SOD) and malondialdehyde(MDA) changes in the spinal cord were observed at the 24 h, 48 h and 72 h after modeling. RESULTS: The hind limb locomotor func-tion of the rats in group A was completely recovered. The hind limb locomotor function of rots in group C and group B improved at 24, 48 and 72 h, with significant improvement at 24 h (P<0.05) and significant difference between 48 and 72 h (P<0.01). In group A, the tissue structure was well preserved, the motor neurons bodies and the nuclcolus were clear and the cytoplasm was stained deeply and uniformly. In group B, the nerve tissue showed multifocal leukoencephaiopathy bleeding and a number of nerve cells disappeared. The nucleus became small because of pycnosis and vacuole appeared around nerve cells at 72 h. In group C, the injury of the nerve tissue was mild, the cells appeared swelling and the cytoplasm was homogeneous. Only a few nucleus became small and few vacuoles appeared around nerve cells at 72 h. SOD activity of spinal cord tissue was high but MDA content was low in group A. The SOD activity of group B reduced obviously and it was significantly different compared with that in group A (P<0.01). SOD activity of group C increased and the MDA content reduced obviously, significantly different compared with those in spinal cord injury group (P<0.01). CONCLUSION: Resveratrol observably promotes the functional recovery of spinal cord after injury and effectively protects the spinal cord from injury.%目的:建立Allen's脊髓模型,探讨白藜芦醇(Res)对大鼠脊髓损伤(SCI)的保护效果.方法:将24只SD大鼠随机分为假手术组(A组),脊髓打击损伤组(B组),Res组(C组),每组8只.A,B组根据自制改良的Allen's装置制备脊髓急性打击损伤动物模型.脊髓打击损伤后即刻腹腔注射Res100 mg/kg,观察并检测3组大鼠SCI后24,48,72 h 3个不同时间点后肢运动功能评分变化及脊髓组织病理学、超氧化物歧化酶(SOD)活性、丙二醛(MDA)含量改变.结果:术后A组大鼠功能完全恢复.B组和C组大鼠后肢运动功能24,48,72 h 3个时间点均有所改善.但24 h时差异明显(P<0.05);48,72 h时差异更为显著(P<0.01).A组神经组织结构完好,神经元轮廓清楚,核仁清晰可见,胞质均匀深染.脊髓打击损伤后72 h B组神经组织多灶性出血并出现广泛的神经元坏死,尼氏体溶解消失,核固缩变小.C组神经组织结构损伤轻微,细胞轻度肿胀,胞质均匀.术后A组脊髓组织SOD活性升高,而MDA含量则处于较低水平.B组SOD活性明显降低,MDA含量明显升高,与A组相比较差异显著(P<0.01).c组与B组相比较,SOD活性明显升高,MDA含量明显降低,差异具有统计学意义(P<0.01).结论:Res能明显促使SCI后的功能恢复,对SCI具有一定的保护作用.
