间接酶联免疫法检测鲑鳟鱼类传染性造血器官坏死病病毒

摘要

Objective To establish an indirect enzyme-linked immune assays (ELISA) for the identification of infectious hematopoietic necrosis virus (IHNV). Methods An experimental rabbit was immunized to produce anti-IHNV serum with IHNV-G protein. To assess the titer and specificity of the anti-serum, the ELISA test was used. Further, PCR was used to verify the above ELISA experimental result. Results The titer of anti-serum of IHNV-G and IHNV reached to 1:32000 and 1:16000, respectively, moreover, the cross-reaction with other virus was 0%, and ELISA results was with 98%accordance rate with the method of PCR. Conclusion The indirect-ELISA of this research has a good specificity and sensitivity, and it is suitable for rapid detection and epidemiological investigation of IHNV infection in trout.%目的：建立快速检测传染性造血器官坏死病病毒(IHNV)的间接酶联免疫法。方法以传染性造血器官坏死病毒G蛋白(IHNV-G)为抗原,免疫实验白兔,制备兔抗IHNV免疫血清,通过ELISA对兔抗血清进行效价及特异性评估,并通过 PCR 方法进行验证。结果应用间接 ELISA 技术,检测到抗血清对 IHNV-G 和IHNV的效价分别达到1：32000和1：16000,与其他对照病毒无交叉反应。与PCR检测结果符合率达到98%。结论本文建立的间接酶联免疫法方法具有良好的灵敏性和特异性,为 IHNV 的检测提供了快捷、高效的技术手段。
