清肺口服液对人喉癌细胞感染呼吸道合胞病毒后ICAM-1表达水平的影响

             

摘要

目的 探讨清肺口服液对人喉癌细胞(Hep-2)感染呼吸道合胞病毒(RSV)后细胞间黏附分子-1(I-CAM-I)表达水平的影响.方法分时段以清肺口服液含药血清,利巴韦林含药血清及空白血清干预RSV感染的体外培养的Hep-2细胞,48h时及72h后收集各组细胞上清,采用酶联免疫吸附试验法(ELISA法)测定各组人喉癌细胞ICAM-I表达变化.结果48h时及72h后病毒模型组ICAM-1的表达明显高于清肺口服液Ⅰ、Ⅱ、Ⅲ组及利巴韦林Ⅱ、Ⅲ组;空白血清组与病毒模型组无显著差异.清肺口服液Ⅰ、Ⅱ、Ⅲ组与利巴韦林Ⅱ、Ⅲ组比较,无显著差异.清肺口服液Ⅰ、Ⅱ、Ⅲ组之间比较,无显著差异.72h后各组ICAM-1的表达与48h相比,无显著差异.结论清肺口服液可降低Hep-2感染RSV后ICAM-1的过度表达,减轻炎症反应,提前给药更可获得良好的效果,这可能是清肺口服液抗病毒的机制之一.%Objective:To investigate the effects of Qingfei Oral Liquid on expression of Intercellular adhesion molecules (ICAM)-l in Human laryngoeareinoma epithelial cells (Hep-2)infected with Respiratory Syncytial Virus (RSV). Methods: Hep-2 cells in vitro culture were intervened respectively with serum containing Qingfei Oral Liquid,ribavirin and blank serum. In these 3 groups,intervention was conducted before,simultaneously or after infecting with RSV (I , II , Ⅲ ).48 hours and 72 hours later cell supernatants were collected respectively. Changes of expression ICAM-1 were detected by enzyme linked immunosorbent assay (ELISA). Results: After 48 and 72 hours of cultivation,expression levels of ICAM-1 in virus control group were significantly higher than those in Qingfei Oral Liquid groups (I , II , Ⅲ ) or Ribavirin group ( II , Ⅲ ) . No significant difference was found between the blank serum control groups ( II , Ⅲ ) and the virus control group. There was no remarkable difference between the Qingfei Oral Liquid groups (I , II , Ⅲ ) and the Ribavirin groups ( II , Ⅲ ). Differences among the Qingfei Oral Liquid groups ( I , II , Ⅲ) were no significance. Expression levels of ICAM-1 within the Qingfei Oral Liquid group (I , II , Ⅲ) were slightly different after 48 or 72 hours of cultivation. Conclusion: Qingfei Oral Liquid can decrease the over-expression of ICAM-1 of Hep-2 cells infected with RSV so as to relieve inflammation. Furthermore, better effects can be achieved when it is given in early stage of RSV infection. These conditions may be one of the antiviral mechanisms of Qingfei Oral Liquid.
