清气化痰汤对急性肺损伤小鼠水通道蛋白1表达的影响

         

摘要

目的 探讨清气化痰汤治疗急性肺损伤(ALI)的机制,为临床治疗ALI开辟新的治疗措施提供理论和实践方向.方法 24只昆明小鼠随机分为空白对照组、模型对照组、清气化痰汤组,观察6h后测定肺湿/于重比值(w/d),并采用HE染色观察炎症变化,RT-PCR检测AQP1 mRNA、肿瘤坏死因子α(TNF-α)mRNA、白细胞介素1β(IL-1β)mRNA的表达变化.结果 模型对照组小鼠的肺部炎症反应显著,以中性粒细胞浸润为主,伴明显的肺泡充血、水肿,清气化痰汤组肺组织炎症、充血明显改善,模型对照组的w/d与其他3组比较,差异有统计学意义;与空白对照组相比,模型对照组AQP1 mRNA的表达显著降低,而清气化痰汤组的AQP1 mRNA的表达显著高于模型对照组.与空白对照组相比,模型对照组TNF-αmRNA的表达显著升高,而清气化痰汤组的TNF-α mRNA的表达显著低于模型对照组.IL-1β mRNA表达的变化与TNF-α mRNA的变化相似,肺损伤炎症减轻.结论 清气化痰汤可通过上调AQP1的表达,减轻肺损伤时肺水肿,可能是通过抑制炎症因子TNF-α、IL-1β的表达实现的.%Objective :To explore the mechanism of Qingqihuatan on the treatment of acute lung injury (ALI) in mice induced by lipopolysaccharide by studying changes of expression of aquaporin1 (AQP1) and to provide a new clinical method in treating ALI. Methods:24 mice were randomly divided into 3 groups, the saline control group (NS) ,the LPS group(ALI) and the Qingqihuatan Decoction group(QT). Wet/dry mass ratio(W/D) and lung tissue histopathological changed were observed at six hours. AQP1 mRNA 、TNF-αmRNA and IL-1βmRNA were assayed by RT-PCR. Results: Histological tissue showed that extensive lung inflammation were seen in the ALI group ,which manifested by accumlation of significant numbers of neutrophils,accompanied by marked pulmonary edema and hermorrhage.Compared with other groups,the difference of the W/D mass ratio of ALI group was not signficant.Compared with NS group, AQP1mRNA expression in ALI group was decreased significantly,while AQP1mRNA expression in Qingqihuatan group was higher than that in ALI group. Compared with NS group,TNF-αmRNA expression in ALI group was increased significantly,while TNF-αmRNA expression in Qingqihuatan Decoction group was lower than that in ALI group. The expression of IL- l βmRNA was similar to that of TNF-αmRNA. Conlusions: Qingqihuatan Decoction may have beneficial effects on ALI by regulating AQP1 expression in the lungs of rats,which are related with the inhibition of IL-1βand TNF-α.
