艾灸头部组穴对VD大鼠脑内VEGF及其受体Flt-1、Flk-1 mRNA表达的影响

         

摘要

Objective:To observe the expressions of VEGF,flt-1,Flk-1mRNA within the hippocampous and cortex of the vascular dementia (VD) rats and to study the mechanism of angiogenesis of the treatment by moxibustion.Methods:Wistar health elderly male rats was selected.4-vessle occlusion was used to prepare the vascular dementia model.The model rats was dIvided into 3 groups by the digit schedule such as the moxibustion group,the western medicine group,the model group.The sham operated group was set to contrast.the moxibustion group rats treated by moxibustion on the point of baihui(GV20),shenting(GV14) and dazhui(GV24).The western medicine group rats was treated with Aniracetam for 2 courses.step-down avoidance test,nerve behavior test and nervial behavior scores were implemented before the treatment and after the 2 courses treatment,respectively.And test the expression level of VEGF,flt-1,Flk-1mRNA within the hippocampous and cortex of the rats after the treatment in RT-PCR ways.Results:After 2 courses treatment,the test scores in latent period,error times and nervial behavior scores showed that the moxibustion group and the western medicine group had significant difference compared with model group.The moxibustion group got better than the western medicine group,which explain that the moxibustion group and the western medicine group had their own treatment effect in curing VD but the moxibustion group had more effective.Statistics of the level of VEGF,flt-1,Flk-1mRNA that the VD rats hippocampus and cortex positive cells expressed showed that the moxibustion group had significant difference compared with model group but the western medicine group had no significant difference.the moxibustion group got the higher level of VEGF,flt-1,flk-1 mRNA than western medicine group too.These all showed that the moxibustion was more effective in increasing the positive cells expression within the VD rat hippocampus and cortex,and the western medicine had no similar effect in curing VD.Conclusion:Moxibustion has the effect in improving behavioral score and memory performance in VD rats,and it maybe exist the mechanisms of control expression level of VEGF,flt-1,Flk-1 mRNA within the hippocampous and cortex.In particular,the moxibustion raise the key factor VEGF,flt-1 Flk-1mRNA expression,and promote angiogenesis in the vital parts,and ultimately stimulate mechanisms of repairation of brain nerve injury.%目的 观察艾灸头部组穴治疗血管性痴呆(VD)大鼠皮层、海马内VEGF及其受体Flt-1、Flk-1 mRNA表达,研究艾灸调控VD脑内血管新生机制.方法 选取Wistar雄性老年大鼠,四血管阻断法制备大鼠VD模型,电脑随机数字表法随机分为艾灸组、西药组、模型组,另设假手术组对照.艾灸组取百会、大椎、神庭穴艾灸治疗,西药组茴拉西坦灌胃共2个疗程.神经行为学评分并结合跳台仪检测治疗前、第2疗程后学习记忆成绩;RT-PCR检测各组大鼠皮层、海马内VEGF及其受体Flt-1、Flk-1 mRNA的表达水平.结果 经2个疗程治疗后,动物跳台实验的潜伏期、错误次数,以及神经行为学评分,艾灸组、西药组与模型组均有非常显著差异,表明艾灸、西药在提高VD大鼠潜伏期,降低VD大鼠错误次数以及对神经行为学评分影响均有明显疗效,且艾灸组明显优于西药组.与模型组相比,艾灸组皮层内VEGF及其受体Flt-1 mRNA的表达有明显差异,海马内VEGF及其受体Flk-1 mRNA的表达有显著差异;而西药组仅显示海马内VEGF或皮层Flt-1表达上调,没有观察到同一脑区VEGF及其受体同步明显表达.结论 艾灸在改善VD大鼠学习记忆成绩、神经行为学评分的同时,通过调控VEGF及其受体Flt-1、Flk-1 mRNA的表达水平,促进了要害部位的血管生成,并最终激发了脑内神经的损伤修复机制.
