益气养阴通络法对兔腹主动脉球囊损伤术后VEGF、bFGF、TGFβ1表达的影响

摘要

目的 观察具有益气养阴、活血通络功效的中药欣舒注射液对兔腹主动脉球囊损伤术后VEGF、bFGF、TGFβ1表达的影响,探讨其防治PCI术后再狭窄的机制.方法 将新西兰大耳白兔随机分为正常组、模型组、欣舒注射液组、阿托伐他汀组.正常组未经手术及用药,其余3组行手术造模.2周后,实验动物麻醉后,采集腹主动脉标本,光镜观察兔腹主动脉,免疫组化方法检测兔腹主动脉VEGF、bFGF、TGFβ1蛋白表达,Western Blot印迹分析检测兔腹主动脉VEGF、bFGF、TGFβ1蛋白表达.结果 兔腹主动脉球囊损伤后,腹主动脉VEGF蛋白表达虽较正常组高,但与正常组比较无显著差异,腹主动脉bFGF、TGFβ1蛋白表达明显升高.中药组、西药组可增加VEGF蛋白表达,中药组、西药组可减少bFGF、TGFβ1蛋白表达.结论 欣舒注射液具有防治PCI术后再狭窄的作用,这可能与其增加腹主动脉VEGF表达,降低bFGF、TGF-β1表达的作用有关.%Objective: To observe the VEGF、bFGF and TGFβ1 expression of rabbits'abdominal aorta after injured by sacculus proprius,under the influence of Xinshu injection,a kind of Chinese herbal medicine,which has effect of supplementing Qi and nourishing Yin;promoting blood circulation to remove meridian obstruction. Also, to investigate the mechanism of Xinshu injection preventing and curing the restenosis after PCI operation. Methods: The new-Zealand big ear white rabbits were divided into 4 groups by random, the normal group (A), the model group (B), the Chinese herbal medicine-Xinshu injection group (C)and the Atorvastatin group (D). The A group hasn't yet done any surgical operation or uses any medicine,where as B,C,D group were done with some surgical operations to build a mode. After 2 weeks,these animals were anesthetized. Then collected the specimen of abdominal aorta,observed under the light microscope,used the immunity method and Western Blot blotting analysis to detect the expression of the protein VEGF、bFGF and TGFβ1. Results: After injured by sacculus proprius , the expression of VEGF protein in abdominal aorta were higher, but there was no significant variance compared with the normal group,the expression of abdominal aorta protein bFGF、TGFβ1 ascended obviously.Both TCM and western medicine could lead to the ascending of the expression of protein VEGF,and both TCM and western medicine could lead to the descending of the expression of protein bFGF、TGFβ1. Conclusion: Xinshu injection can prevent and cure the restenosis after PCI operation, and its effect is possibly related to the mechanism that Xinshu injection can elevate the protein VEGF expression , while can degrade the expression of bFGF、 TGF-β1.
