黄芩黄酮提取影响因素及与黄连配伍后对黄酮溶出的动态影响

             

摘要

目的 探讨黄芩提取不同因素对黄酮类化合物溶出的影响,并观察黄芩与黄连不同比例配伍后对黄酮溶出的动态影响,探讨两者配伍的意义.方法 以芦丁为对照品,建立黄酮含量测定方法;考察加水量、颗粒大小、提取时间、提取次数等对黄芩总黄酮提取的影响;测定黄芩与黄连不同比例配伍后不同时间点黄酮的含量.结果 芦丁标准曲线y=0.0139x+0.0651 (R2=0.9993),在7.68~53.76 μg/mL质量浓度范围内与吸光度呈良好的线性关系;加水量、颗粒大小、提取时间、提取次数均对黄芩总黄酮的提取具有较明显的影响;黄芩与黄连以不同的比例配伍后明显改变了黄芩中黄酮的溶出,其中当黄芩与黄连3∶1时,在同时间点黄酮溶出最高.结论 考察的提取因素分析可为黄芩黄酮的工业化提取提供参考,并从化学角度探讨了黄芩与黄连不同比例配伍后成分变化的规律及意义.%Objective:To investigate the effect of different factors on the dissolution of flavonoids in Scutellaria baicalensis Georgi,to observe the dynamic effects of different proportions of baicalin and Rhizoma Coptidis on the dissolution of flavonoids,and to explore the significance of the compatibility.Methods:Rutin was used as reference substance to establish a method for the determination of flavonoids.The effects of water volume,particle size,extraction time and extraction times on the extraction of total flavonoids from Scutellaria baicalensis Georgi were investigated.The content of flavone was determined at different time points in different compatibility of Scutellaria baicalensis Georgi and Coptis chinensis.Results:The standard curve of rutin y=0.0139x+0.0651 (R2=0.9993),in 7.68~53.76 g/mL range of concentration and absorbance showed a good linear relationship;water content,particle size,extraction time and extraction times had an obvious effect on the extraction of total flavonoids of Scutellaria baicalensis;compatibility of Scutellaria baicalensis Georgi and Coptis chinensis in different proportions significantly changed the dissolution of Flavonoids from Scutellaria baicalensis Georgi.when 3∶1,the highest flavonoids dissolution reached at the same time.Conclusion:The analysis of extraction factors can provide reference for the industrial extraction of flavonoids from Scutellaria baicalensis,and discuss the law and significance of the composition change of different proportions of Scutellaria baicalensis and Coptis chinensis from the chemical point of view.
