女贞叶的直接电离质谱研究

         

摘要

Ambient ionization (AI) techniques allow the analysis of high complex sample in their native state with minimal sample pretreatments.Obvious advantages of simplicity,speed and versatility have been brought to mass spectrometry analysis with the analytes directly ionized without matrix clean-up.AI-MS was employed to directly fingerprint the phytochemicals in Ligustrum lucidum Ait leaf.By adding some methanol and applying high voltage on the "Ⅴ" shape sample,a variety of phytochemicals such as sugars,alkaloids and flavonoids were extracted from the sample,and electrospray occurred at the tip of the sample carrying microdroplets containing the dissolved analytes for mass spectrometric interrogation.The metabolism information during the vegetation progress of Ligustrum lucidum Ait leaf were preliminary investigated by comparing the phytochemicals patterns of the half-growth and the full-growth leaves.Experimental results provide useful information for the pharmaceutical research and clinical applications of Ligustrum lucidum Ait leaf.%直接电离质谱技术(AI-MS)可以在无需样品预处理的条件下直接分析复杂基体样品,为实现复杂基体样品的简单、快速分析提供了可靠的途径.利用AI-MS对女贞叶进行直接质谱分析,快速获得了不同生长阶段女贞叶的质谱指纹谱图,初步研究了女贞叶生长过程中化学成分的变化情况.实验表明,女贞叶舍有较丰富的钾、糖类、单黄酮、双黄酮等有益成分.为开展女贞叶的植物生理考察及药用价值开发提供了一种简单、快速的质谱分析方法,也简明讨论了女贞叶作为慢性心肌炎等疾患预防用药的物质基础,为女贞叶药材的临床应用提供了科学依据.
