病毒性肝炎时肝B1-a细胞对肝脏保护作用的实验研究

             

摘要

Objective; To detect the dynamic changes of mice liver B-1a cells and to investigate the effect of B-1a cells in murine fulminant viral hepatitis. Methods; The percentage of B cells and B-1a cells among liver lymphocytes of A/J mice and Balb/cj mice were detected via flow cytometry, while the changes after infection of MHV-3 were also detected. Further, the liver B-1a cells were isolated from liver lymphocytes of Balb/cj mice. Afterwards, they were transplanted into Balb/cj mice via tail vein injection. The histopathological characteristics were observed after infected with MHV-3 for 72 hours. Results; The percentage of B cell in A/J mice was higher than that of Balb/cj mice (43. 6% ±2. 1% vs 15. 6% ±3. 3% , P <0. 01) , while the percentage of B-1a cell in these two type mice had no significance different (6.38% ±2.13% vs 5.84% ±1.32% , P>0.05). After infected with MHV-3, the percentage of B-1a cell was increased ( 10.80% ±2.82% , P < 0. 05) in A/J mice, while decreased in Balb/cj mice. Furthermore, after infected with MHV-3 for 3 days, the liver necrosis were slight in mice transplanted with B-1a cells than those of control groups. Conclusion; The liver intrinsic B-1a cells could protect the liver from immune injury induced by viral infection.%目的:检测小鼠感染鼠肝炎病毒3型( MHV-3)后肝脏B-1a淋巴细胞亚群变化,探讨B-1a细胞在病毒性肝炎中的作用.方法:通过流式细胞术检测MHV-3诱导的耐受型小鼠及引起暴发性肝炎敏感小鼠肝脏中B细胞、B-1a细胞的基础水平,以及在感染后不同时间点的比例变化.磁珠分选出未感染敏感小鼠肝脏B-1a细胞并过继给敏感小鼠,观察感染MHV-3病毒72 h后小鼠肝脏的病理组织学特征.结果:未感染时MHV-3耐受型小鼠肝脏中B细胞明显高于敏感型小鼠(43.6%±2.1% vs 15.6%±3.3%,P<0.01),B-1a细胞在耐受型小鼠略高于敏感型小鼠(6.38%±2.13% vs 5.84%±1.32%,P>0.05);感染后耐受型小鼠B-1a明显升高至(10.80%±2.82%,P <0.05),而敏感型小鼠呈现下降趋势.过继B-1a细胞的敏感小鼠肝脏坏死程度明显低于其它细胞组及PBS对照组.结论:肝脏固有B-1a细胞对MHV-3引起的病毒性肝炎具有保护性作用.
