尿α1微球蛋白对妊娠期大鼠子痫前期的预测价值

             

摘要

[目的]探讨尿α1微球蛋白(α1-MG)水平对妊娠期大鼠子痫前期(PE)的预测价值.[方法]26只SPF级SD雌性大鼠确认妊娠后随机分为PE模型组及正常妊娠组,每组13只.自妊娠d11起,PE模型组给予尾静脉注射血管紧张素Ⅱ-1型受体拮抗剂(AT1-AA),正常妊娠组尾静脉注射等量生理盐水,共注射7 d.妊娠d10起采用动物无创血压测试仪测量孕鼠自开始尾动脉收缩压;妊娠d8、d12、d16、d20收集两组孕鼠尿液,采用速率免疫散射比浊法检测两组孕鼠尿α1-M G含量;临分娩前剖宫取仔,比较两组孕鼠的平均仔鼠数、畸形率、死胎率,测仔鼠体重、胎盘湿重.[结果]妊娠12~21 d,PE模型组尾动脉收缩压较正常妊娠组显著增高(P <0.05),PE模型组尾动脉收缩压在妊娠12~18 d显著增加,且相邻两天比较差异有统计学意义(P <0.05);PE模型组在妊娠期d12、d16、d20尿α1-M G含量显著高于正常妊娠组(P <0.05),PE模型组组内尿α1-M G含量两两相比,差异均有统计学意义(P <0.05);PE模型组平均仔鼠数、仔鼠体重及胎盘湿重显著低于对照组(P <0.05),PE模型组畸形率及死胎率均显著高于正常妊娠组(P <0.05).[结论]AT1-AA诱导的PE大鼠模型中,PE大鼠出现妊娠期高血压,尿α1-M G水平较高,产仔质量差,推测尿α1-M G水平对大鼠PE发病有重要预测价值.%[Objective]To evaluate the predictive value of urine α-1 microglobulin(α1-MG)level for pre-eclampsia(PE)during gestational rats.[Methods]A total of 26 female SD rats under SPF condition were con-firmed to be pregnant.These SD female rats were randomly divided into the PE model group and the normal preg-nancy group,with 13 in each group.From the 11th day of pregnancy,rats in PE model group received AngiotensinⅡ-type 1 receptor antagonist(AT1-AA)while mice in normal pregnancy group received equal volume of saline by tail vein injection for 7 days.The tail artery systolic pressure was measured by animal's noninvasive blood pres-sure tester from the 10th day of pregnancy.The urine of pregnant rats of the two groups was collected on the 8th, 12th,16th,and 20th day of pregnancy.The concentration of urine α1-MG in the two groups was detected by rate immunoassay.Caesarean section was performed before pregnancy end.The average number of newborn rats,the rate of deformity and stillbirth rate were compared between the two groups.The weight of newborn rats and wet weight of placenta were measured.[Results]At 12 to 21 days of gestation,the systolic blood pressure of the tail artery in the PE model group was significantly higher than that of the normal pregnancy group(P <0.05).The systolic pressure of the tail artery in the PE model group increased significantly from the 12th to the 18th day of gestation,and there was a statistically significant difference between the two adjacent days(P <0.05).The level of α1-MG in urine of d12,d16and d20during the gestational period was significantly higher in the PE model group than in the normal pregnancy group(P <0.05).The difference between the urinary α1-MG levels in the PE model group was statistically significant(P <0.05).The average number and weight of newborn rats and wet weight of placenta in PE model group were significantly lower than those in the normal pregnancy group(P <0.05).The de-formity rate and stillbirth rate in the PE model group were significantly higher than those in the normal pregnancy group(P < 0.05).[Conclusion] Pregnancy hypertension,higher levels of urinary α1-MG,and poor quality of newborn rats are observed in the AT1-AA induced PE rat model.So,it is speculated that the detection of urine α1-MG level has important predictive value for preeclampsia in rats.
