三七总皂苷对急性肺损伤相关炎性因子及抗氧化因子的调控及保护作用

         

摘要

目的 研究三七总皂苷对SD大鼠失血性休克型急性肺损伤相关炎性因子及抗氧化因子的调控作用和肺损伤保护作用.方法 SD大鼠24 只,随机分为3 组.A组为假手术组,只进行相应的手术操作;B组和C组制作失血性休克模型,造成急性肺损伤,然后在20 min 内复苏,C 组复苏时应用三七总皂苷.测定动脉血浆中肿瘤坏死因子-α(TNF-α)含量、支气管肺泡灌洗液(BALF)中巨噬细胞(PAM)和中性粒细胞(PMN) 总数、BALF 中蛋白含量和肺组织中超氧化物歧化酶(SOD)含量;计算肺组织含水率;光镜下观察肺组织形态学改变.结果 与A 组比较,复苏后B、C 组TNF-α含BALF 中的蛋白含量和PMN 总数、肺组织含水率明显升高,SOD 含量明显降低.但C 组上述指标的变化程度均低于B 组.光镜下见B 组肺组织间质水肿、细胞浸润明显,与之相比C 组肺组织以上改变明显减轻.结论 三七总皂苷对SD大鼠的失血性休克型急性肺损伤具有保护作用.%Objective To study the protective effects of Total saponins of Panax Notoginseseng on SD rats hemorrhagic shock a-cute lung injury . Methods Twenty-four SD rats were divided into 3 groups randomly which named group A, B and C respectively. Group A was non-operation group, while hemorrhagic shock models were performed in both group B and group C, and resulted in ALI, then the shock rabbits were resuscitated in 20 min. The Total saponins of Panax Notoginseseng was used in group C during resuscitation. The arterial blood was taken in different time to determine the plasma tumor necrosis factor-α( TNF-α ) concentration. The amount of pulmonary alveolarmacrophages ( PAM ) and polymorphonuclear leukocyte ( PMN ) in bronchoalveolar lavage fluid ( BALF ) were counted at-the end of experiment. At the same time, the protein contents in BALF and superoxide dismutase( SOD ) contents in lung tissue were measured, and the water ratio of lung tissue was calculated. The histological forms of lung were examined through the light microscope. Results Compared between group A and group B, TNF-α concentration, the protein contents and the amount of PAM in BALF were increased more obviously after resuscitation. In group C the level of all indexs above was lower than that in group B. In contrast to the group A, SOD contents of lung tissue were lower in groupB, with a high level of water ratio in lung tissue. In group C the SOD contents were higher than those in group B, while the water ratio was reduced. Lung tissue edema and neutrophil accumulation can be observed by microscope both in group B and group C, but the changes in the later were mollified. Conclusion Total saponins of Panax Notoginseseng has the protective effects on ALI which resulted from experimental hemorrhagic shock in SD rats .
