氟西汀对CUMS模型组大鼠血浆miR-34a miR-134 miR-143表达的影响

             

摘要

目的 探讨血浆miR-34a、miR-134、miR-143成为抑郁行为严重程度和药物疗效的客观评估指标的可能性.方法 将40只雄性SD大鼠分为4组,干预组、模型组、模型+干预组、对照组各10只,采用慢性不可预见性应激的方法建立抑郁模型,采用旷场实验评价大鼠行为. 结果 应激前4组大鼠各项实验指标比较差异无统计学意义 (P>0.05);而应激后,模型组和模型+干预组大鼠24 h饮用1%糖水消耗量及体质量、水平运动格子数与垂直运动格子数较应激前显著减少,排便次数较应激前显著增多(P<0.05或0.01),对照组和干预组则无显著变化(P>0.05).本研究未检测到miR-143的表达水平.与对照组相比,模型组大鼠血浆miR-34a表达增强(P<0.05),miR-134表达下降(P<0.05);与模型组相比,模型+干预组、干预组大鼠血浆miR-34a表达下降(P<0.05),miR-134表达增强(P<0.05).结论 应激抑郁模型小鼠糖水代谢消耗下降,动力及探索活动减少,而氟西汀可增加糖水消耗动力及探索活动.miR-143在血浆中基本不表达,血浆miR-134与miR-34a有可能成为抑郁症的生物学标记物,值得进一步研究.%Objective To explore the possibility of plasma miR-34a, miR-134 and miR-143 becoming objective indexes evaluating severity of depressive behaviors and drugs' efficacy.Methods Forty male SD rats were divided into intervention, model, model + intervention, and control group of 10 ones each, depressive models established with chronic unpredictive stress method, and rats' behaviors evaluated using the Open Field Test (OFT).Results There were no significant differences in all experimental indexes before stress among 4 groups (P>0.05);after stress 24 hour drinking 1% syrup consumption, body mass, the frequencies of horizontally and vertically crossing a grid line reduced more significantly and defecation increased compared with pre-stress in model and model + intervention group (P<0.05 or 0.01) and had no notable changes in control and intervention group (P>0.05).The miR-143 expression levels weren't detected in the study.Plasma miR-34a expression reinforced in model compared with control group (P<0.05) and lowered in model + intervention and intervention compared with model group (P<0.05).Plasma miR-143 expression lowered in model compared with control group (P<0.05) and reinforced in model + intervention and intervention compared with model group (P<0.05).Conclusion Syrup metabolic consumptions lower, dynamic and exploring activities reduce in stress depression model rats, and fluoxetine could increase syrup metabolic consumptions as well as dynamic and exploring activities.The miR-143 doesn't express in plasma basically, plasma miR-134 and 34a are possible to become biological markers of depression and deserve further study.
