肾母细胞瘤黑色素抗原基因-1mRNA的表达及临床意义

摘要

Objetive To investigate the mRNA expression of melanoma antigen-1 mRNA in Nephroblastoma and its'clinical significance. Methods MAGE-1 mRNA expression were determined by RT-PCR in 32 cases of nephroblastoma tissues and 10 cases of paracancer renal tissues and 6 cases of renal tissue without tumour. The relations about MAGE-1 mRNA expression were also analyzed in nephroblastoma' s different clinical stage and different pathology type. Results Some degree of MAGE-1 mRNA expression was present in all samples. The MAGE-1 mRNA level was 1.4-fold higher in tumour tissues than in paracancer renal tissues(t = 0. 853 ,P > 0. 05 ) and 2. 58-fold higher than in renal tissue without tumour( t = 2. 35 , P < 0. 05 ). We also en-duce that MAGE-1 mRNA level do not correlate with pathology type and clinical stage in nephroblastoma(P > 0. 05). Conclusion MAGE-1 mRNA in nephroblastoma is expressed so high that it has important significance for immunotherapy and prognosis of nephroblastoma.%目的 探讨小儿肾母细胞瘤中黑色素抗原基因-1(MAGE-1) mRNA的表达及意义.方法 用逆转录多聚酶链反应(RT-PCR)方法 检测32例肾母细胞瘤组织、10例瘤旁组织、6例无瘤肾组织的MAGE-1 mRNA的表达情况,并分析MAGE-1 mRNA的表达与肾母细胞瘤的病理分型、临床分期之间的关系.结果 所有标本均有不同程度的MAGE-1 mRNA表达,表达程度按肿瘤组织、瘤旁组织、无瘤组织依次降低.肿瘤组织是瘤旁组织的1.40倍(t=0.853,P>0.05),是无瘤对照组的2.58倍(t =2.35,P<0.05).MAGE-1 mRNA在肾母细胞瘤组织中的表达与病理分型及临床分期均无关(P>0.05).结论 MAGE-1 mRNA在肾母细胞瘤组织中呈高表达,提示MAGE-1 mRNA可望成为肾母细胞瘤免疫治疗的靶抗原.
