Objective To explore the mechanism of Survivin antagonistic peptide on the proliferation of C6 glioma cells through investigating the inhibitory effect of Survivin antagonistic peptide on the proliferation of C 6 glioma cells .Methods MTT assay was used to detect the inhibitory effect of Survivin antagonist peptide on the proliferation of C 6 rat glioma cells .Immunohistochemical method was used to detect the expression changes of Caspace -3 in experimental and control groups with or without Survivin antagonistic peptide in C 6 rat glioma cells .Results MTT results showed that with the inhibited effect of C 6 glioma cells proliferation was strengthen in compare with the concentration increase of Survivin antagonistic peptide .What is more , the inhibitory rate rised in a dose-and time-dependent manner .Treatment with 20μg/ml Survivin antagonist peptide for 72 h, the inhibitory rate was 62%, which was exhibited in a dose-dependent manner .Immunohistochemistry showed that with the increase of Survivin antagonistic peptide concentration , further the expression of Caspace-3 in the nuclei of C6 rat glioma cells was gradually increased .Conclusion Survivin antagonistic peptide could up-regulate the expression of caspase-3 expression in the nucleus , further promoting cell apoptosis , and may play a strong inhibitory effect on C 6 glioma cells .%目的:观察Survivin拮抗肽对C6胶质瘤细胞生长抑制作用,探讨Survivin拮抗肽对C6胶质瘤细胞增殖的作用机制。方法用MTT法检测Survivin拮抗肽对C6大鼠胶质瘤细胞增殖抑制影响;免疫组化法检测Survivin拮抗肽作用前后实验组和对照组C6大鼠胶质瘤细胞中Caspace-3的表达水平。结果 MTT结果显示随着Survivin拮抗肽浓度的增加,对C6胶质瘤细胞的增殖抑制作用逐渐增强,且随浓度及作用时间延长抑制率上升,20μg/ml Survivin拮抗肽作用72 h后,抑制率为62%,呈剂量依赖性。免疫组化显示随着Survivin拮抗肽浓度的增加,Caspace-3在C6大鼠胶质瘤细胞细胞核的表达逐步增加。结论 Survivin拮抗肽可使Caspase-3在细胞核中的表达增多,从而促进细胞凋亡,可能对C6胶质瘤细胞发挥较强的抑制作用。
