PURPOSE: New strain of Pseudomonas fluorescens LS20 (KCTC 8912P) producing siderophore is provided which inhibits the growth of plant pathogenic zhizobacteria and promotes the growth of plant. CONSTITUTION: Siderophore productive microorganism is isolated from ginseng roots by plating diluted solution of ginseng roots and soil on chrome azurol S (CAS) blue agar plate. API 20NE diagnosis strip is used to identify Gram negative bacteria. Productivity of siderophore of isolated bacteria is tested using spectrophotometric analysis of CAS- hexadecyltrimethylammonium bromide complex and total activity of siderophore is examined by CAS shuttle analysis. The type of siderophore is analysed by arrow phenol acid method. Petri plate analysis is used to detect antagonism properties of isolated microorganism on the plant pathogenic fungus, Fusarium solani. The antifungal activity of siderophore produced from isolated microorganism is examined by inoculation of microorganism on the iron-deficient KB medium. The plant growth-promoting activity of isolated mocroorganism is verfied by inoculation of Fusarium infected Phaseolus vulgaris.
