Toll样受体4和血管内皮生长因子与烟雾暴露大鼠肺动脉重塑的相关性研究

         

摘要

Objective To explore the correlation of Toll-like receptor 4 (TLR4) and vascular endothelial growth factor(VEGF) with pulmonary artery remodeling in rats exposed to smoke.Methods Forty eight male healthy rats of SPF level were randomly divided into S0 group(with no smoke exposure),S1 group(with smoke exposure for 1 month),S2 group(with smoke exposure for 2 months) and S3 group (with smoke exposure for 3 months) with 12 rats in each group.Hematoxylin and eosin(HE) staining was used to observe the morphological changes of pulmonary artery.The proportion of pulmonary artery wall area/total vascular area(WA%)and pulmonary artery wall thickness/vascular external diameter(WT%)were measured by medical image analysis system.The expression of TLR4 and VEGF in pulmonary artery were measured by immunohistochemical staining.Then the correlation of TLR4 and VEGF with WA% and WT% were analyzed.Results WA%,WT% were aralyzed,TLR4 and VEGF in S1 group,S2 group and S3 group were higher than those in S0 group significantly (P < 0.05),and all were directly proportional to the time of smoke exposure.TLR4 was positive correlated with WA% and WT% (r =0.893,0.902,P <0.01).VEGF was positive correlated with WA% and WT% (r =0.887,0.896,P < 0.01).TLR4 was positive correlated with VEGF(r =0.935,P <0.01).Conclusion TLR4 and VEGF are correlated with pulmonary artery remodeling in rats with smoke exposure,and TLR4 is correlated with VEGF.The upregulation of TLR4 and VEGF levels may be one of the mechanisms of pulmonary artery remodeling in rats exposed to smoke.%目的 探讨Toll样受体4(TLR4)和血管内皮生长因子(VEGF)与烟雾暴露大鼠肺动脉重塑的相关性.方法 SPF级雄性健康大鼠48只,随机分为S0组(无烟雾暴露)、S1组(烟雾暴露1月)、S2组(烟雾暴露2月)、S3组(烟雾暴露3月),每组各12只大鼠.在苏木素-伊红(HE)染色下观察肺动脉形态学变化,并测量肺动脉管壁面积/血管总面积(WA%)和肺动脉壁厚度/血管外径(WT%).免疫组织化学染色法检测肺动脉TLR4和VEGF的表达.分析TLR4、VEGF与WA%、WT%及TLR4与VEGF的相关性.结果 S1组、S2组和S3组WA%、WT%、TLR4和VEGF均高于SO组,且与烟雾暴露时间呈正比,差异均有统计学意义(P<0.05).TLR4与WA%、WT%均呈正相关(r=0.893,0.902,P均<0.01);VEGF与WA%、WT%均呈正相关(r=0.887,0.896,P均<0.01);TLR4与VEGF呈正相关(r=0.935,P<0.01).结论 TLR4和VEGF与烟雾暴露大鼠肺动脉重塑相关,而且TLR4与VEGF也相关,TLR4及VEGF过表达可能是烟雾暴露大鼠肺动脉重塑的机制之一.
