芒果苷对脑性瘫痪模型大鼠神经运动功能及皮质区神经细胞凋亡的影响

             

摘要

Objective To explore the effect of mangiferin (MAN)on the nerve motor function and cortical neuronal apoptosis in cerebral palsy rats. Methods Cerebral palsy model was made by ligating the left common carotid artery. 60 SD rats were divided into 5 groups:the sham group,Model group,MAN (10 mg/ kg)group,MAN (25 mg/ kg)group and MAN (50 mg/ kg)group. BBB score was valued in the 1st,15th and 30th day atfter surgery. In the 30th day,all brain tissue was collected,and immunohistochemical staining was made to value the caspase - 3 and Bax protein expression,meanwhile,the relative protein expression of Bax and Bcl - 2 were detected by western blotting. Results BBB score showed that there was no significant difference between the MAN group and model group (P > 0. 05),and in the 15th and 30th days,BBB score in MAN group was markedly higher than that in model group (P 0.05),第15和第30天,芒果苷组BBB评分显著高于模型组,并呈剂量依赖趋势(P<0.05);免疫组织化学染色显示,芒果苷组caspase-3、Bax蛋白平均光密度显著低于模型组,并呈剂量依赖趋势(P<0.05);Western blot-ting结果显示,芒果苷组Bax蛋白灰度值显著低于模型组(P<0.05),而Bcl-2蛋白灰度值显著高于模型组,并呈剂量依赖趋势(P<0.05).结论 芒果苷能显著改善脑性瘫痪大鼠神经运动功能,抑制脑皮质区神经细胞凋亡,并其作用呈剂量依赖性.
