High pressure freezing(HPF)can preserve excellent cellular structures in a near-native state with samples in glasslike amorphous ice quickly. Next freezing substitution(FS)steps are processed at low temperatures. We applied HPF and FS technology on yeast Saccharomyces cerevisiae and observed the ultrastructure. The goal of this study is to provide some technological references for researchers that want to investigate at the ultrastructural level in yeasts.%高压快速冷冻技术可以使生物样品快速冷冻固定,其组织、细胞结构接近于生理状态,有效减少了样品制备过程中化学试剂对细胞结构的破坏,并能够观察到细胞器近似自然状态下的超微结构.本文应用高压冷冻和冷冻替代技术进行酵母细胞的电镜样品制备,观察到了清晰的细胞壁、细胞核、液泡、线粒体及内质网等亚细胞器的超微结构,为酵母细胞的电镜样品制备提供了更多的方法和手段.
