小鼠肾集合管发育中的细胞凋亡与水通道蛋白2的表达

         

摘要

Abstract Objective To observe the expression of AQP-2 and cell apoptosis in collecting ducts of mice during development, and to discuss the relationship between AQP-2 and cell apoptosis. Methods Immunohistoche-mistry,electron microscope,TUNEL and stereology were used to detect the expression of AQP-2 and cell apoptosis in the collecting ducts of mice at E17 d,E18 d,Nl d,N7 d,N14 d,N21 d and N40 d. Results Cell apoptosis could be seen at N1 d in collecting ducts and reached the peak at N7 d. The expression of AQP-2 was weak at E17 d but strongest at N7 d,with the surface density reaching the maximum (0.044 1±0.001 8 μm-1) as well.From N21 d to N40 d, both cell apoptosis and expression of AQP-2 were unchanged. Conclusion The results suggest that AQP-2 has some effect on cell apoptosis and paly an important role in the development of collecting ducts.%目的 观察小鼠肾集合管发育过程中细胞凋亡与水通道蛋白2(AQP-2)的表达,探讨两者的关系及在集合管发育过程中的作用.方法应用免疫组织化学、电镜、TUNEL染色及体视学方法对胚龄(E)17,18d及生后日龄(N)1,3,7,14,21,40d小鼠肾集合管中的细胞凋亡与AQP-2的表达进行观察和分析.结果细胞凋亡出现于N1d的集合管,N7 d达到最高峰.AQP-2从E17 d的胎鼠开始微弱表达,到N7 d表达强度最强,面密度值也达到最大水平,为(0.044 1-0.001 8)μm-1.N21 d至N40 d细胞凋亡和AQP-2的表达都无明显变化.结论细胞凋亡在集合管的发育过程中起重要的作用,AQP-2对细胞凋亡有一定的影响.
