Aβ25-35对PC12细胞凋亡蛋白Bcl-2和Bax表达的影响

         

摘要

目的::观察Aβ25-35对PC12细胞凋亡蛋白Bcl-2和Bax表达的影响。方法:采用10μmol/L Aβ25-35与PC12细胞共孵育48h建立阿尔茨海默病细胞模型,对照组细胞不加Aβ25-35。采用Hoechst 33258核染色法检测PC12细胞的凋亡情况,免疫细胞化学染色法检测PC12细胞Bcl-2和Bax蛋白的表达情况。结果:与对照组相比,实验组细胞Bcl-2的表达明显降低,细胞凋亡率、Bax蛋白的表达和Bax/Bcl-2比值明显升高(P<0.05)。结论:Aβ25-35诱导PC12细胞凋亡的机制可能与下调Bcl-2表达、上调Bax表达有关。%[ABSTRACT]Objective:To explore the effects of amyloid beta-protein (25-35) on Bcl-2 and Bax expression in PC12 cell. Methods:Alzheimer’s disease cells model was made by culturing the PC12 cells with 10μmol/L Aβ25-35 for 48 hours. Hoechst 33258 method was used to detect the apotosis of PC12 cells, and immunocytochemical stain was used to detect the expression of Bcl-2 and Bax protein in PC cells. Results:Compared with control group, the Bcl-2 expression in PC12 cells of experimental group decreased obviously, while the apoptosis rate, Bax expression and Bax/Bcl-2 increased obviously (P<0.05). Conclusions:The mechanisms of PC12 cells apoptosis induced by Aβ25-35 may be related to down-regulation of Bcl-2 expression and up-regulation of Bax expression.
