紫杉醇在常压反相层析柱上的纯化

         

摘要

Taxol, a diterpene obtained from Taxus spp., has significant anti-cancer activity and unique mechanism of anti-cancer action. Due to the very low contents in plant and cell culture with large amounts of impurities present, it is difficult to obtain high recovery at high purity of taxol in various methods, and the HPLC process is always essential. A typical separation and purification process would result in less than 60% recovery if 98% of purity were obtained. In order to prepare taxol in large scale and reduce the expense of production, after the initial separation step, the C18-silica gel reversed phase chromatography at normal pressure was used to purify taxol from Taxus yunnanensis. The operation conditions of C18-silica gel chromatography were optimized. The capacity of loading and the increasing recovery of taxol were performed. The crude sample of taxol could be separated and purified by this technology without HPLC process, the purity of taxol reached more than 98% from 25% with the recovery of 85%.%在C18-硅胶柱上常压反相层析法对从云南红豆杉浸膏中初分离的紫杉醇进行了纯化精制。对C18-硅胶层析纯化紫杉醇的操作条件进行了优化,并对层析进料量及紫杉醇回收率的有效提高进行了研究。采用本工艺分离精制紫杉醇粗品,可使紫杉醇含量从25%左右提高到98%以上,回收率大于85%。
