选取红冠桉茎段作为外植体,建立了无菌再生体系;对外植体选择、不定芽增殖、生根诱导及移栽各阶段的关键因子进行了探讨.结果表明:(1)茎尖为较适宜的外植体;(2)最佳增殖培养基为M+6-BA 1.5 mg/L+NAA0.2 mg/L+VB2 6.0 mg/L;(3)最佳的生根培养基为1/2M+IBA 1.5 mg/L+VB2 6.0 mg/L,生根率达96.3%;(4)发酵过的椰糠与黄心土按3:2混合而成的基质为适宜的移栽基质,移栽成活率达81.15%.本研究首次利用茎段建立了红冠桉无菌再生体系,为今后的研究及大规模生产奠定了技术基础.%Stem segment of Corymbia ptychocarpa was collected as explant, and key factors for each regeneration stage, including explant selection, adventitious bud proliferation rooting induction and transplanting were tested. The results show that: (1)The stem tip was the best explant; (2) The adventitious shoots were subcultured and multiplied on an improved MS medium containing 1.5 mg/L 6-BA, 0. 2 mg/L NAA and 6. 0 mg/L VB2; (3) Rooting medium with 1/2 MSmedium supplied with 1.5 mg/L IBA and 6. 0 mg/LVB2 induced a rooting rate up to 96. 3%; (4) The transplant medium was the mixture of coconut husk and yellow soil by ratio of 3 to 2. The survival rate of transplants reached 81. 15%.
