中药复方通腑颗粒对抗生素相关性肠炎大鼠肠黏膜微生态屏障损伤的保护作用

         

摘要

Objective To invesligale the effecls of Tongfu granules on inleslinal microflora barrier funclion in rals with anlibiolic-associaled colilis. Tongfu granules which is a kind of Chinese Iradilional medicine. The presenl sludy aimed Lo invesligale prolecling effecl of the medicine on impaired inleslinal mucosa, and furlher sludy effecls of Dahuang and Houpo which are the Iwo main components of Tongfu granules. Methods Eighly male rals were divided inlo len groups. Afler making the models of Lincomycin induced anlibiolic-associaled colilis, blood samples of rals were laken for measuring endoloxin. The colonic conlenls of rals were collected lo cullure four floras; bacillus acidi laclici, bacillus bifidus, enlerococci bacillus coli. The pathological change of inleslinal mucosa was observed using lighl microscope. Results The body weighl of the model group was significantly decreased, a significant difference was shown between Houpo and Tongfu granule in weighl. The endotoxin in rals of trealment group was decreased. Bui insignificant difference was shown among Dahuang, Houpo and Tongfu granule groups in endoloxin. There was a significant difference (P <0. 05) belween Tongfu granule and model group after Ireatmenl. The number of bacillus acidi laclici, bacillus bifidus, enlerococci bacillus coli increased. The scores of the lesions showed a significant difference between Tongfu granule and Dahuang. Conclusion There was injury in inlestinal mucosal barrier in anlibiotic-associaled colitis rals. Chinese Traditional medicine Tongfu granules Dahuang and Houpo had protective effects on the small inteslinal barrier. They can decrease the endoloxin level of antibiolic-associated colilis rats. They also could recover the inteslinal flora. Tongfu granule was betler than Dahuang and Houpo.%目的 探讨中药复方通腑颗粒对抗生素相关性大鼠的肠道微生态屏障的保护作用,进一步比较通腑颗粒2种主要成分大黄、厚朴在此过程中的作用.方法 采用SD 大鼠80只,以4 g/kg盐酸林可霉素灌胃制备抗生素相关性肠炎模型,将大鼠按数字表法随机分为大黄组、厚朴组、通腑颗粒组和模型组,模型制作成功后开始给药.观察各组体质量变化、内毒素情况.取大鼠结肠内容物进行乳酸杆菌、双歧杆菌、肠球菌、大肠杆菌4种菌的培养与计数.同时取各组大鼠空肠组织做常规固定、染色,光学显微镜观察.结果 模型组大鼠体质量明显下降,治疗后通腑颗粒组大鼠较大黄组、厚朴组体质量明显增加.通腑颗粒组、大黄组与厚朴组内毒素浓度降低,但3组之间差异无统计学意义.通腑颗粒、大黄与厚朴均具有调节肠道菌群作用,通腑颗粒组疗效更优.模型组小肠黏膜病理损伤评分明显高于其他治疗组,治疗后通腑颗粒组与大黄组比较,小肠黏膜病理损伤明显降低.结论 抗生素相关性肠炎大鼠存在肠黏膜屏障损伤;中药通腑颗粒、大黄、厚朴可以有效减轻肠黏膜损伤,纠正大鼠的肠道菌群紊乱,中药通腑颗粒疗效更优.
