电针对大鼠脊髓损伤后notch1、ps1表达的实验研究

     

摘要

Objective To discuss the protective mechanism of electroacupuncture in rats after spinal cord injure (SCI). Methods All rats were randomly divided into the model group, blank group, medicinal group and electroacupuncture group. The model of SCI was established by using the modified Allen's method. The injured spinal tissue was taken after 6 hours, 1 day and 7 days respectively for extracting total RNA. The expressions of notchl and psl mRNA were detected by using reverse transcription polymerase chain reaction (RT-PCR). Results The expressions of notchl and psl mRNA all decreased significantly in the electroacupuncture group and medicinal group after SCI for 6 hours, 1 day and 7 days. Compared with the model group, the expressions of notchl and psl mRNA decreased in the medicinal group and electroacupuncture group (P<0.05). The comparison between the medicinal group and electroacupuncture group showed that the expressions of notchl and psl mRNA were higher a little in the electroacupuncture group but without significant difference (P > 0. 05 ). Conclusion Notch signal passway plays a very important role in the growth, development, differentiation and apoptosis of cells. The activation of notch signal can irreversibly accelerate NSC differentiation to gliacytes. Electroacupuncture has a significant inhibitory effect on the expressions of notchl and psl after SCI.%目的 探讨电针治疗对大鼠脊髓损伤( SCI)后的保护机理.方法 大鼠随机分为模型组、空白组、药物组、电针组,用改良Allen's打击法制成脊髓损伤模型,分别于损伤后6h、1d、7d取损伤局部脊髓组织,提取总RNA,经RT-PCR反应,测定损伤后脊髓组织内信号通路基因(notch1)、早老素1( ps1) mRNA的表达.结果 脊髓损伤后,电针组和药物组均能显著降低脊髓损伤6h、1d、7d后脊髓组织中notch1、ps1 mRNA的表达.与模型组相比较,药物组和电针组notch1、ps1 mRNA表达均降低,并都有显著性差异(P<0.05);药物组与电针组相比较,电针组notch1、ps1 mRNA表达略高于药物组,但无显著性差(P>0.05).结论 notch信号通路,在细胞的生长、发育、分化及凋亡中起着十分重要的作用,notch信号的激活可加速并不可逆地促进神经干细胞(NSC)向胶质细胞分化,电针治疗对大鼠脊髓损伤后notch1、ps1表达具有明显的抑制作用.
