五子衍宗丸含药血清对大鼠睾丸支持细胞活力的影响

         

摘要

Objective To observe the influence of the medicated serum of Wuzi Yanzong Wan on vitality of sertoli cells of testis in rats. Methods The original strain of sertoli cells of rat testis was cultured and medicated serum samples of Wuzi Yanzong Wan were prepared in high dose and low dose. The high-dose and low-dose serum samples were then divided into 20% group, 10% group, 5% group and blank group, and cultured together with original strain of sertoli cells of rat testis. The influence of Wuzi Yanzong Wan on the vitality of sertoli cells was observed by using methyl thiazolyl tetrazolium test (MTT) at 5 time points including 6 h, 12 h, 24 h, 48 h and 72 h. Results At the time points of 6 h, 12 h, 24 h, 48 h and 72 h, the vitality of sertoli cells increased in 20% group and 10% group along with the prolong of time, reached the highest at 24 h and decreased gradually after that time point. Conclusions The medicated serum of Wuzi Yanzong Wan has some improving effect on the vitality of sertoli cells, and the vitality will be the highest when 20% medicated serum and sertoli cells are cultured together for 24 hours.%目的 观察五子衍宗丸含药血清对大鼠睾丸支持细胞活力的影响.方法 培养大鼠睾丸原代支持细胞,制备高、低剂量五子衍宗丸含药血清,高、低剂量组合药血清又进一步分为20%、10%、5%和空白血清4个中药血清浓度培养基,并将不同浓度五子衍宗丸含药血清分别和大鼠睾丸原代支持细胞共同培养,选取6、12、24、48、72 h5个时间点,MTr法观察在不同时间点时五子衍宗丸对支持细胞活力的影响.结果 培养6、12、24、48、72 h后,随着时间的增加,20%及10%浓度血清组细胞活力亦随之增加,在24 h达到最高值,然后细胞活力随着培养时间的增加逐渐下降.结论 五子衍宗丸含药血清对支持细胞的活力有一定的促进作用,且在高剂量组20%浓度血清培养基和支持细胞共培养24 h后,支持细胞活力最高.
