紫外线抵抗相关基因在二烯丙基二硫诱导人白血病 K562细胞自噬中的作用

         

摘要

目的:观察紫外线抵抗相关基因(UVRAG)在二烯丙基二硫(DADS)诱导白血病 K562细胞自噬中的作用。方法以 K562细胞为细胞模型,利用透射电镜观察细胞的超微结构自噬体的形成;采用吖啶橙染色和荧光显微镜观察酸性囊泡的形成以检测 DADS 诱导 K562细胞自噬;RT-PCR 检测自噬相关基因UVRAG mRNA 的表达水平。结果透射电镜显示:与空白组和溶媒组比较,DADS(40 mg·L -1)处理组可观察到胞质内出现大量自噬体。吖啶橙染色荧光显微镜下可见到各 DADS 处理组 K562细胞胞浆中的酸性囊泡较空白组和溶媒组明显增多。RT-PCR 结果显示,DADS 作用 K562细胞12 h 后,各 DADS 处理组UVRAG mRNA 表达水平均高于空白组和溶媒组。结论 DADS 能诱导 K562细胞自噬,其机制可能与上调UVRAG mRNA 表达有关。%Objective To observe the value of ultraviolet radiation resistance-associated gene(UVRAG)in the diallyl disulfide(DADS)induced autophagy of K562 cells. Methods K562 cells were chosen for this study. Their ultrastructure was observed by transmission election microscopy. The formation of autophagic vacuoles were observed by fluorescence microscope after acridine orange staining. RT-PCR was used to detect the expression levels of UVRAG mRNA. Results Compared with the blank group and the menstruum group,large amount of autophago-somes were observed in the cytoplasm of K562 cells treated with DADS(40 mg·L - 1 );amount of autophagic vacu-oles observed in all the DADS treatment groups were observed;the UVRAG mRNA expression levels were higher in all the DADS treatment groups after DADS treatment for 12 h(P < 0. 05). Conclusion DADS can induce autoph-agy of K562 cells,it may be related to the up-regulation of UVRAG mRNA expression.
