蒙药童格勒格-1对高脂大鼠的降脂作用及其机制研究

         

摘要

目的:探讨蒙药童格勒格-1(TGLG -1)对高脂大鼠血脂的调节作用及对大鼠脂肪组织肿瘤坏死因子-α(TNF -α)表达的影响。方法:以高脂饲料喂养大鼠1个月建立高血脂动物模型,用 TGLG -1灌胃,6周后测定大鼠血脂和脏器系数的改变,并观察肝脏的形态变化;用免疫组织化学法检测脂肪组织 TNF -α的表达及脂肪细胞数目的变化。结果:高脂大鼠血清总胆固醇(TC)、甘油三酯(TG)水平高于对照组( P <0.01),且肝组织出现明显的脂肪变性,脂肪细胞体积增大且数目减少,说明模型制作成功;用蒙药 TGLG -1治疗6周后,大鼠血清 TC、TG 水平降低,高密度脂蛋白胆固醇(HDL - C)水平升高( P <0.01),肝组织脂肪变性有所缓解;免疫组织化学检测结果显示,高脂大鼠脂肪组织TNF -α表达高于其它3组,而蒙药 TGLG -1组表达最低( P <0.01)。结论:蒙药 TGLG -1具有较好的降低高脂大鼠血脂和预防高脂血症的作用,其降脂作用可能与 TNF -α有关。%Objective:To probe into the adjustive effect of TGLG - 1 on the level of serum lipids and expression of TNF -α in the adipose tissues of hyperlipidemic rats. Methods:The model of hyperlipidemic rats was established with one - month high - lipid diet. Using full - automatic bio - chemical analyse instrument,the level of serum lipids,organ coefficient and the changes ofliver morphology in the rats in each group were determined 6 weeks after gastro - perfusion of TGLG - 1. The expres-sion of TNF - α in adipose tissues and the changes in the number of lipocytes were detected by immunohistochemical method. Results:The levels of serum total cholesterol(TC)and triglyceride(TG)in the rats in hyperlipidemia model group were signifi-cantly higher than those in the control group( P < 0. 01),with adipose degeneration significant in hepatic tissue,lipocytes en-larged and the number decreased,which suggested that the model was successful. The levels of TC and TG significantly dropped and the level of high - density lipoprotein(HDL - C)significantly rose( P < 0. 01)6 weeks after the treatment with TGLG - 1, with adipose degeneration alleviated in hepatic tissue. The results of immunohistochemistry showed that the expression of TNF -α in adipose tissue was more significant in hyperlipidemic model group than that in the other three groups( P < 0. 01),with the expression of TNF - α least significant in TGLG - 1 group( P < 0. 01). Conclusion:Mongolia drug TGLG - 1 could remarkably lower the serum lipid level in hyperlipidemic rats and prevent hyperlipidemia,whose hypolipidemic effect may be related to TNF - α.
