碱提取法从虎杖中分离白藜芦醇的方法研究

         

摘要

The method of the extraction and separation of resveratrol with alkali solution has been studied in this experiment, in which we have adjusted the pH value by hydrochloric acid to precipitate. The effects of the raw material granularity, kind and dosage of alkali, acid dosage and the time of soaking have been studied in this ex-periment by single factor. The optimum conditions of resveratrol alkali extraction and acid precipitation were as follows:filtering the raw material through the 40 mesh sieve, regulating the distilled water to pH 10 with KOH, soaking the material for 10 hours, boiled it l h under a constant rate and even agitation stir, finally regulating the filtered liquid to pH 3 by 1 mol/L hydrochloric acid, and precipitation. The crude extract really contains resvera?trol after detected by UV and IR spectrum. It has been demonstrated that it is a good technique with simple pro?cess and a safe operation, so it is an effective separation method.%实验对白藜芦醇采用无机碱性水溶液进行提取、然后选择适宜的pH值进行沉降,完成对白藜芦醇的分离.采用单因素实验,对原料粒度、无机碱的种类及用量、酸的用量、浸泡时间等因素进行探究,确定了碱提取酸沉淀方法分离白藜芦醇的最佳条件:原料粒度为过40目筛;提取溶剂为氢氧化钾调节蒸馏水pH值为10;浸泡时间为12 h;恒速、均匀搅拌,加热煮沸1 h;抽虑,沉淀试剂为1 mol/L的盐酸调节pH值为3,静置充分沉淀.所得到的粗提物经过紫外、红外光谱检测,确实含有白藜芦醇.表明碱提取酸沉淀的分离方法工艺简单,操作安全,是行之有效的分离方法.
