HPLC法测定贺兰山东麓酿酒葡萄中苹果酸含量

         

摘要

[Objective] The study aimed to optimize the chromatographir conditions of HPLC for detecting the malic acid in the wine grape. [Method] With 6 grape varieties from the planting base in the Yuqueanying in the eastern foot of Helan Mountain as the test materials, the malic acid content in the grape juice was detected by HPLC whose chromatographic conditions were as follows; Zorbax SB CIS column (5.0 μm, 250. 0 mm ×4.6 mm), column temperature of 30 ℃ ; with 0. 02 mol/L potassium dihydrogen phosphate solution ( pH = 2. 75 ) as the mobile phase, sampling volume of 10.0 μl,detected wavelength of 210 nm,flow rate of 0.6 ml/min. The investigation on the linear relationship, the tests of the precision and recovery, and the inspection of the repeatability and stability were conducted. [ Result ] The malic acid contents in the samples of 6 grape varieties was in 2.00-4.50 g/L. RSD was 0.3% , linear correlation coefficient R2 was 1.000, and the average recovery of assay was 98. 9%. The chromatographic conditions of the method could completely separate each grape sample and had the characteristic of high accuracy, small error, good repeatability, simple operation. [Conclusion] The method of detecting the malic acid content in different grape grapes from the eastern foot of Helan Mountain was established.%[目的]优化HPLC测定酿酒葡萄中的苹果酸的色谱条件.[方法]以来自贺兰山东麓玉泉营葡萄种植基地的6个葡萄品种为试样,用HPLC法测定葡萄汁中的苹果酸含量,其色谱条件为:Zorbax SB-C18柱(5.0μm,250.0 mm × 4.6 mm);柱温30℃;以0.02 moL/L磷酸二氢钾水溶液(pH =2.75)为流动相,进样量10.0μl,检测波长210 nm,流速0.6 ml/min,并进行了线性关系的考察,精密度和加标回收率试验以及重复性和稳定性检验.[结果]6个葡萄品种样品中的苹果酸含量范围在2.00 ~4.50 g/L,RSD为0.3%,线性相关系数R2=1.000,加标平均回收率为98.9%.该方法的色谱条件可将每种葡萄样品进行很好地分离,具有准确度高、误差小、重现性好、简便快捷的特点.[结论]建立了用HPLC法测定贺兰山东麓不同品种葡萄中苹果酸含量的方法.
