枣树叶片蛋白质提取及SDS-PAGE单向电泳条件优化

         

摘要

[目的]对枣树叶片蛋白质提取方法及其SDS-PAGE单向电泳条件进行优化.[方法]对枣树5种蛋白质提取方法和影响SDS-PAGE单向电泳的因素进行比较研究.[结果]改良丙酮沉降法提取出的蛋白质杂质较少,条带数量多且清晰,同时通过研究确定了一套适用于枣树蛋白检测的SDS-PAGE凝胶制备及电泳方法.[结论]该研究为进一步从分子生物学的角度研究枣树与枣疯病植原体的互作机制提供了技术的基础.%[ Objective ] The aim was to optimize the protein extraction method of Chinese jujube leaves and its 3DS-PAGE single dimensional electrophoresis conditions. [ Method] 5 protein extraction methods and factors affecting the SDS-PAGE single dimensional electrophoresis were compared and optimized in Chinese jujube. [Result] The protein extracted by modified acetone settling process has less impurities and more limpid strap number, meanwhile, a set of Chinese jujube protein detection method of SDS-PAGE gel preparation and its electrophoresis was I-dentified. [ Conclusion] This study prepared the technical ground for the further research of interaction mechanism between Chinese jujube and jujube witches broom phytoplasma from the perspective of molecular biology research.
