三七茎基总皂苷的回流提取工艺研究

         

摘要

[目的]确定三七茎基总皂苷含量测定方法,优化其回流提取工艺.[方法]采用香草醛-高氯酸法测定三七茎基总皂苷含量的方法,并以总皂苷含量为指标,优选回流法提取三七茎基总皂苷的工艺条件.[结果]在0.008 3 -0.0496 mg/ml浓度范围内,人参皂苷Rg1浓度与吸光度的线性关系良好(R=0.999 4),平均回收率为99.3%(n=4),RSD为3.4%.最佳提取工艺条件为:料液比1∶10(g/ml)、乙醇浓度60%,浸泡时间12 h,提取3次,每次2h.[结论]该含量测定方法简便、准确、重现性好;提取工艺经济、合理,值得进一步推广.%[ Objective] To optimize the methods for content determination and backflow extraction technology of total saponins in stem base of Panax notoginseng ( Burk) F. H.. [ Method] The content of total saponins was determined by the methods of vanillin-perchloric acid and the extraction technology was optimized by the content of total saponins. [ Result] Panax saponin Rg1 presented good linear relationship over the range of 0.008 3 to 0. 049 6 mg/ml (r =0.999 4). The average recovery was 98.6% with the RSD of 3. 54% (n=4) respectively. The optimal solvent baclcflow extraction conditions were as follows:P. notoginseng stem base was soaked for 12 h in 60% ethanol at the solid-liquid rati-o of 1:10 g/ml and extracted for 3 times for 2 h each lime. [Conclusion] This method is simple and feasible,highly accurate and precise. The extraction process is economical, reasonable and deserved popularizing all the more.
