方秆蕨中多糖含量的测定

     

摘要

[目的]确定方秆蕨多糖的最佳提取工艺,测定方秆蕨中的多糖含量.[方法]采用水提醇沉法提取方杆蕨多糖,研究料液比、提取时间、提取温度对多糖提取率的影响.采用苯酚-硫酸法,以葡萄糖为标准品,蒸馏水为空白对照,于490nm波长处分别测定对照品和供试品溶液的吸光度,按标准曲线计算方秆蕨中的多糖含量.[结果]方秆蕨多糖的最佳提取条件为料液比1:40(g/ml)、温度80℃、时间2.5h.[结论]方秆蕨多糖的含量为2.5%.%[Objective] To determine the optimum condition of extracting polysaccharide and the content in Glaphytopteridopsis erubescent (Wall. ) . [Method] Polysaccharide was extracted from Glaphylopteridopsis erufescens (Wall. ) by decoction and alcohol sedimentation technique. The effect of solid to liquid ratio, extracting time and temperature on extracting polysac-charide from Glaphylopteridopsis erubescens (Wall. ) was studied. The content of polysac-charide in Glaphylopteridopsis erubescens (Wall. ) was determined by the phenol-sulferic acid method, with the glucose was as reference standard and the distilled water as the blank control, the absorbance of the control and test solution was determined at 490 nm and the content of polysaccharide was calculated by using the standard curve. [ Result] The optimum extraction conditions were determined as follows,solid to liquid ratio,1:40(g/ml); extracting time,2. 5 h; temperature,80 T. [ Conclusion] The po-lysaceharide content could reach 2. 5% under the optimum condition.
