极早熟李品种'红美丽'的组织培养与快速繁殖

     

摘要

[Objective] To establish the system of tissue culture and rapid propagation of early matured plum cultivar"Red Beauty".[ Meth-od] Semi-lignified young shoots were selected as explants, the effects of basal medium composition on the initial growth of axillary bud were in-vestigated.In vitro shoots were selected as materials, the effects of basal medium composition and the concentration of plant growth regulators on proliferation of tube seedling and rooting of tube seedling were examined.[ Result] Basal medium QL evidently improved the sprouting per-centage of axillary buds in inducing the initial growth of axillary buds when half lignification stem section was selected as explant.For prolifera-tion, WPM was best, followed by MS and QL.For rooting percentage, there was no significant difference among different auxin concentra-tions.But the higher concentration of 2 mg/L IBA significantly improved the average root number per plantlet than lower concentration of 1 mg/L IBA.The best rooting medium was MS+2 mg/L IBA+0.5 mg/L NAA.Rooting percentage was 83.5%.[Conclusion] This research laid a research and technical foundation for virus-free plantlet production of plum cultivar"Red Beauty"and breed improvement by using bi-otechnology breeding methods.%[目的]建立极早熟李优良品种"红美丽"的组织培养快繁技术体系.[方法]以半木质化的嫩枝茎段为外植体,研究基本培养基对腋芽萌发的影响.以试管苗为试材,研究基本培养基及不同浓度的植物生长调节物质对试管苗增殖及生根的影响.[结果]以半木质化的嫩枝茎段为外植体进行腋芽的启动培养,QL比MS明显提高腋芽萌发率;试管苗增殖培养以WPM最佳,其次为MS和QL;不同生长素浓度上的生根率无显著差异,但较高浓度(2 mg/L)IBA上的平均单株生根数显著高于较低浓度(1 mg/L)IBA上的.最佳生根培养基为MS+2 mg/L IBA+0.5 mg/L NAA,生根率为83.5%.[结论]该研究为"红美丽"李无病毒苗木的生产及利用生物技术育种方法进行品种改良奠定了研究和技术基础.
