铁皮石斛多倍体诱导研究

     

摘要

[Objective] The research aimed to study the polyploid induction of Dendrobium officinale.[Method]Dendrobium officinale capsule were used as materials to induce round bulb,the effects of different concentrations of NAA and 6-BA on the proliferation of round bulbs of Dendrobium officinale were studied.The effects of different concentrations of colchicines and treatment time on the induction of polyploid Dendrobium officinale were studied by using colchicine as an inducer.[Result]When the concentration of NAA was 1.5 mg/L,the concentration of 6-BA was 2.0 mg/L,the effect of the proliferation of round bulbs of Dendrobium officinale was the best.The colchicine concentration was lower,treatment time was shorter,the survival rate was higher,the induction rate was relatively low;with the increase of colchicine concentration and treatment time prolonged,the induction rate increased,but increased mortality.[Conclusion] When the concentration of colchicine was 0.15% and the treatment time was 48 h,the survival rate was up to 40%,and the induction rate was the best,which was 30%.%[目的]研究铁皮石斛的多倍体诱导.[方法]以铁皮石斛蒴果为材料,诱导萌发出铁皮石斛圆球茎,研究不同浓度的NAA、6-BA对铁皮石斛圆球茎增殖的影响;以秋水仙素作为诱导剂,研究不同浓度秋水仙素、处理时间对多倍体铁皮石斛诱导的影响.[结果]NAA浓度为1.5 mg/L、6-BA浓度为2.0 mg/L时,铁皮石斛圆球茎增殖效果最好;秋水仙素浓度越低、处理时间越短时,成活率越高、诱导率相对较低;随着秋水仙素浓度增大,处理时间延长,诱导率升高,但死亡率增大.[结论]秋水仙素浓度在0.15%、处理时间为48 h时,成活率达40%,诱导率为30%,效果最好.
