高氯酸钠急性暴露对斑马鱼胚胎发育及氧化应激的影响

     

摘要

It was chosen perchlorate as exposure compound ,the zebrafish embryo as a test organism to study the effects of acute toxicity and oxidative stress of perchlorate ( PER) to zebrafish embryos .The acute toxicity was processed by the statistical data of surviving rate ,malformation rate,heart rate and hatching rate for zebrafish eggs/larvae;the effects of oxidative stress was processed by SOD、CAT, MDA and Gpx , and relative mRNA expression levels of genes ( Mn-Sod、Cu-Zn-Sod1 ) .The results indicated that PER induced significantly developmental toxicity to zebrafish embryos ,which demonstrated by high death rates and malformation rate in the high concentrations group .Exposure 2000 mg/L PER induced significantly oxidative stress in embryos /larvae,which demonstrated by the activities of SOD and MDA were significantly increased ( P <0.01 ), whereas Gpx levels were significantly decreased (P<0.01);in the same time,the transcription of genes (Mn-Sod、Cu-Zn-Sod1 ) related to oxidative stress were significantly up-regulation ( P <0.01 ).These results indicated that PER could induce acute developmental toxicity and oxidative stress in zebrafish embryos /larvae.%实验选择高氯酸钠( PER)作为染毒化合物,将斑马鱼胚胎作为受试生物,研究了高氯酸钠对其的发育过程的急性毒性及氧化应激效应.通过对不同质量浓度高氯酸钠(250、500、1000、1500、2000 mg/L)暴露后胚胎/幼鱼发育指标的统计,考查其急性发育毒性.采用SOD、CAT、MDA以及GSH-px、以及Mn-Sod与Cu/Zn-Sod基因表达的分析,研究了高氯酸钠对斑马鱼胚胎/幼鱼的氧化应激效应.结果显示,PER具有显著地发育毒性,表现为高浓度PER导致胚胎/幼鱼死亡,诱导畸形.同时,2000 mg/L PER暴露显著诱导胚胎/幼鱼产生氧化应激,表现为SOD活性显著增加(P<0.01),MDA含量显著升高(P<0.001),Gpx活性显著降低(P<0.01),以及氧化应激功能基因Mn-Sod与Cu/Zn-Sod表达显著上调(P<0.01).综上可见,PER存在急性发育毒性,能够诱导斑马鱼胚胎/幼鱼产生氧化应激效应.
