电针对偏头痛大鼠5-HT1D受体表达的调控作用研究

             

摘要

目的 研究电针对偏头痛大鼠5-HT1D受体的调控作用,探索电针治疗偏头痛的作用机制.方法 成年雄性SD大鼠50只,随机分为5组:对照组(C组),模型组(M组),单穴组(风池)(EA1组),配穴组(风池+阳陵泉)(EA2组),非穴组(NA组).C组只予以电极安置手术,其余各组在电刺激后予以相应电针治疗.电子von Frey测量大鼠头面部机械痛阈(第0、2、4、6天);实时荧光定量PCR和western blot对偏头痛大鼠三叉神经节(TG)与三叉神经脊束核尾侧亚核(TNC)的5-HT1D受体表达进行检测.结果 在第0天,各组大鼠机械痛阈组间差异无统计学意义;在第2、4和6天,M组大鼠机械痛阈显著低于C组(P<0.001),EA1组和EA2组分别与C组比较差异无统计学意义,NA组与M组差异无统计学意义.M组5-HT1D受体表达显著降低(P<0.001),EA1组与EA2组的5-HT1D受体表达显著高于M组(P<0.001),NA组与M组差异无统计学意义,EA2组的5-HT1D受体表达高于EA1组(P<0.05).结论 电针对偏头痛大鼠TG与TNC部位的5-HT1D受体表达起到上调作用,可能是针刺治疗偏头痛的机制之一.%Objective To explore the mechanism of electroacupuncture in the treatment of migraine, the aim of this study was to learn the effect of electroacupuncture on expression of 5-HT1D receptor in migraine rats. Methods 50 adult male SD rats were randomly divided into five groups: Control group (C), model group (M), electroacupuncture at Fengchi (GB20) group (EA1), electroacupuncture at GB20 and Yangliangquan (GB34) group (EA2), non-acupuncture point group (NA). M, EA1, EA2 and NA were given electronic stimulation to mimic migraine attacks every other day and were treated by eletroacupuncture everyday while C was only given surgery of planting electrodes. On the days 0, 2, 4 and 6, the mechanical pain thresholds of face of each rat were test. The 5-HT1D receptor expression in TG and TNC were detected using real time PCR and Western Blot. Results On day 0, there was no significant difference on mechanical pain thresholds of the face between groups. On the days 2, 4 and 6, mechanical pain thresholds of M was significantly lower than the C group (P<0.001), and there was no significant difference between NA and M. According to the results of real time PCR and western blot, 5-HT1D receptor expression of M was significantly lower than C (P<0.001), EA1 and EA2 had significant higher than M (P<0.001) and no significant difference was observed between NA and M. Moreover, 5-HT1D receptor expression of EA2 was significantly higher than EA1(P<0.05). Conclusion It is possible that upregulate of electroacupuncture on the expression of 5-HT1D receptors in TG and TNC of the migraine rats is one of the mechanisms of the anti-migraine effect of electroacupuncture.
