Objective To investigate the effect of bevacizumab on the lung injury induced by mechanical ventilatioa Methods Eighteen Wistar rats were randomly and equally divided into three groups of A (sham operated), B (mechanical ventilation model) and C( treated with bevacizumab 20 mg/kg before mechanical ventilation). After ventilation, the bronchoalveolar lavage fluid and lung tissues were collected and stored by routine methods. The concentration of vascular endothelial growth factor(VEGF) in the lavage fluid and the content of total protein were determined by ELISA and Brodfard,respectively. The lung capillary permeability was evaluated by Evans blue content and the ratio of dry to wet weight of the lungs. The pathological change of lung tissue was detected by HE staining and the index of lung injury was calculated. Results Compared with group A, the concentration of VEGF in the bronchoalveolar lavage fluid, the lung capillary permeability and the lung injury were all significantly increased in group B(P<0. 05), which were higher than those in group C (P<0.05). Conclusion Bevacizumab can treat and prevent the acute lung injury induced by mechanical ventilation.%目的 研究贝伐单抗对机械通气引起的肺损伤的预防作用.方法 18只Wistar大鼠随机均分为三组:假手术(A)组、机械通气(B)组和贝伐单抗20 mg/kg治疗(C)组.通气结束后,常规取肺泡灌洗液与肺组织,ELISA法检测灌洗液中的血管内皮生长因子(VEGF)浓度,Brodfard法测定蛋白含量,依文氏蓝含量和肺干/湿重比值评价肺毛细血管通透性,HE染色观察肺组织的病理改变并计算肺损伤指数.结果 与A组相比,B组肺泡灌洗液中VEGF浓度、肺毛细血管通透性与肺损伤均显著增加(P<0.05),而C组上述指标均较B组明显降低(P<0.05).结论 贝伐单抗对机械通气引起的肺损伤有一定的预防效应.
