不同参数对低频诊断超声联合微泡辐照下肿瘤血管通透性的影响

摘要

目的 探讨辐照时间、机械指数(MI)和微泡浓度3种参数对低频诊断超声联合微泡辐照下肿瘤血管通透性的影响.方法 2014年11月至2016年1月选取清洁级SD大鼠72只,建立大鼠肝癌模型,并对所有肿瘤组织均进行低频超声联合微泡辐照,对辐照时间、MI值及微泡剂量进行调整,观察肿瘤组织辐照后EB渗出情况.结果 随着微泡剂量的增加(0、60、100、300μ1/kg),肿瘤组织的EB渗出量[(171.50±19.40)、(301.40±15.30)、(372.40±27.40)、(723.10±55.20)μg/g]呈明显上升趋势(P<0.05);随着辐照时间(0、1、5、10 min)的延长,肿瘤组织的EB渗出量[(154.20±19.50)、(372.40±41.50)、(546.70±39.20)、(717.50±55.20) μg/g]呈明显上升趋势(P<0.05);随着MI值(0.0、0.1、0.8、1.3)的增大,肿瘤组织的EB渗出量[(148.90±20.10)、(155.90±30.20)、(311.40±28.78)、(717.50±55.20)μg/g]呈明显上升趋势(P<0.05).结论 MI值、辐照时间及微泡剂量均是影响超声靶向微泡破坏技术治疗的重要参数,其中微泡剂量300μ1/kg联合辐照时间10 min、MI 1.3可最大限度地提高肿瘤组织的血管通透性.%Objective To explore the influence of irradiation time,mechanical index (MI),and microbubble concentration of low-frequency ultrasound and microbubble irradiation on tumor vascular permeability.Methods From November,2014 to January,2016,72 SD rats were selected to establish rat models of liver cancer.The tumor tissues took low-frequency ultrasound and microbubble irradiation;the irradiation time,MI value,and microbubble concentration were adjusted.The EB exudation volume after irradiation in tumor tissue was observed.Results The EB exudation volume in tumor tissue [(171.50±19.40),(301.40±15.30),(372.40±27.40),and (723.10±55.20) μg/g] increased with the dose of microbubble (0,60,100,300 μl/kg).The EB exudation volume in tumor tissue [(154.20±19.50),(372.40±41.50),(546.70±39.20),and (717.50±55.20) μg/g] increased with irradiation time (0,1,5,and 10 min).The EB exudation volume in tumor tissue [(148.90±20.10),(155.90±30.20),(311.40±28.78),and (717.50±55.20) μg/g] increased with MI value (0,0.1,0.8,1.3).Conclusions MI value,irradiation time,and the dose of microbubble are important parameters influencing treatment by UTMD.300 μl/kg microbubble,10 min irradiation,and MI 1.3 can maximize tumor vascular permeability.
