摘要

Objective To study the 5'CpG island methylation patterns of melanoma antigen gene MAGE-A3 , and relative application as tumor marker of hepatocellular carcinoma( HCC). Methods BSP was used for the detection of the methylation /demethylation status of CpG site among the 5'CpG island of MAGE-A3 in genomic DNA from HepG2 cells and white blood cells(WBC)of healthy controls to identify the HCC cells specific demethylation sites of CpG islands. Results Several CpG sites in MAGE-A3 promoters were found with different methylation patterns in HCC cells , as compared to those in WBC of healthy controls , and demethylation patterns of CpG island specific for HepG2 cells were indentified. Conclusion Demethylation pattern of MAGF-A3 5'CpG islands could be used as tumor marker for the early diagnosis of HCC.%目的 研究黑色素瘤抗原基因MAGE-A3 5′端CpG岛异常甲基化状态,探讨其作为肝细胞肝癌(HCC)肿瘤标志物的可行性.方法 利用重亚硫酸盐测序聚合酶链反应(BSP)分析HCC细胞株HepG2和健康对照者来源白细胞基因组中MAGE-A3基因5′端CpG岛异常甲基化状态,寻找HCC细胞MAGE-A3 5′端CpG岛去甲基化位点.结果 HepG2细胞和健康对照者来源白细胞基因组MAGE-A3 5′端CpG岛存在不同的甲基化状态,前者存在特异性去甲基化位点.结论 MAGE-A3 5′端CpG岛去甲基化位点的检测可用于HCC的早期诊断.
