高浓度胰岛素对视网膜M(u)ller细胞VEGF表达的影响

         

摘要

AIM: To study the effect of high concentration insulin on the expression of vascular endothelial growth factor (VEGF) in cultured rabbit retinal Muiler cellsin vitro.METHODS: Muller cells were cultured with insulin ofdifferent concentrations (4× 103, 8× 103, 12× 103 U/L). Immunocytochemistry,in situ hybridization and ELISA were conducted to assay the expression of VEGF in cultured Muller cells in vitro at different insulin concentrations qualitatively and quantitatively.RESULTS: VEGF expression was enhanced obviously by high concentration of insulin.CONCLUSION: Insulin plays an important role in neovascularization of diabetic retinopathy by stimulating the transcription of the VEGF gene in Muiler cells so as to enhance the expression of VEGF protein.%目的:探讨在高浓度胰岛素条件下,体外培养的兔视网膜Muller细胞的血管内皮细胞生长因子(vascular endothelial growthfactor,VEGF)的变化.方法:在不同浓度胰岛素条件下(4,8,12kU/L),体外培养兔视网膜Muiler细胞,采用免疫细胞化学法、原位杂交法,定性测定Muller细胞分泌VEGF的变化,采用ELISA方法,定量测定Muiler细胞分泌VEGF的变化.结果:高浓度胰岛素能明显增强VEGF的表达(P<0.05).结论:高浓度胰岛素可能通过刺激Miiller细胞编码VEGF基因的转录,进而增强VEGF蛋白的表达,而在糖尿病视网膜病变的新生血管生成中发挥重要作用.
