Factors affecting protoplast form ation and regeneration of the Aspergilus niger sp. L-1 from acid rod soil of Longquan M ou tain w ere investigated. The protoplast of stiain L-1, culturod for 30 h in a m inin al m edium coniaining 0 .15 mol/L M gS04 as oanotic slab ilizer, was processed with 0 .3% cellu lase and 0 4% snail enzym e at 30℃ for 120 m inutes. The regeneration frequency of protoplasts could reach up to 11 .3% and cultured for 20 h in a m inim alm edium again . The protoplast of stiain L-1 w as mutated by U V , and then the strain L-H.7 was obtained. The results showed that soluble phosphorus concentra tion in he m edium increased 5.7 tin es, 3 .6 tin es higher than thatof he original stiain. A bility of phosphorus-dissolying of L-17 in acid rod soil increased by 2 .5 tin es than B acillus. M egatherium var. Phosphaticum .J1.%以从龙泉山地区酸性红壤中分离的黑曲霉L-1(Aspergillus niger sp.L-1)为出发菌株,通过研究菌株L-1原生质体的形成和再生条件,发现培养30 h的菌丝体在以0.15 mol/L氯化钾为渗透压稳定剂的基本培养基上经过再次培养20 h后,所得菌丝体用0.3%纤维素酶和0.4%蜗牛酶在30 ℃条件下处理120 min可得大量原生质体,其再生率可达到11.3%.菌株L-1原生质体经过紫外诱变选育后得到L-17,可以使培养基中可溶性磷浓度增加5.7倍,较出发菌株提高3.6倍.L-17在酸性红壤中的解磷能力比解磷巨大芽孢杆菌J1提高2.5倍.
