中温木聚糖酶高产真菌菌株筛选及产酶条件

         

摘要

A β-xylanase producing fungi, Fusarium sp. FLH28, was isolated from soil and humus samples from Xinxiang at 30 ℃ by using basal medium supplemented with xylan as the sole carbon source. The strain FLH28 was capable of growing well at 20-40 ℃; and the optimum growth temperature was 30 ℃. The optimized enzyme production conditions of the strain were as follows, initial pH, 6.0; corncob as carbon source; peptone, yeast extraction and corn steep liquor as nitrogen sources; incubating for 4d at 30 ℃- Under the optimized conditions, the maximum enzyme activity reached to 562.6 U/mL. Optimum pH and temperature of the xylanase was 6.5 and 40 ℃ respectively.%利用以木聚糖作为惟一碳源的基础盐培养基,在30℃培养条件下,从新乡周边土样及腐殖质中筛选到1株β-木聚糖酶高产真菌菌株(Fusarium sp.FLH28).该菌株在20~40℃生长良好,最适生长温度30℃.产酶培养条件优化结果表明,在培养温度为30℃,培养基初始pH 6.0,以玉米芯为碳源,以蛋白胨、酵母粉和玉米浆为有机氮源,发酵4d,木聚糖酶活性达562,6 U/mL,木聚糖酶最适pH和温度分别为6.5和40℃.
