橡胶树胶孢炭疽菌细胞自噬相关基因CgAtg4的克隆与序列分析

摘要

细胞自噬(Autophagy)有助于植物病原真菌渡过营养饥饿、供氧不足等不良环境,有研究发现其与植物病原真菌的致病性相关。利用橡胶胶孢炭疽菌HBCg01全基因组数据库,采用PCR和RT-PCR克隆细胞自噬相关基因CgA tg4(Genebank登录号：KC735125),并对其序列进行分析,研究细胞自噬在胶孢炭疽菌侵染橡胶树过程中的作用。结果表明,CgA tg4的开放阅读框(ORF)为960 nt,编码319个aa序列,预测含有一个由213个aa组成的高度保守的Peptidase C54蛋白酶结构域,与另外19种真菌的Atg4氨基酸序列同源性为38%~87%。推测CgA tg4可能与橡胶树胶孢炭疽病菌的产孢能力、分生孢子萌发以及附着胞膨压的形成相关。%Autophagy can help plant pathogenic fungi survive in nutrition hunger, oxygen hunger and other adverse environ-ment. and It is found to be related with the pathogenicity in some researches. In order to explore the molecular autophagy mechanism of rubber tree Colletotrichum gloeosporioides, the autophagy related gene CgAtg4 (Genebank number:KC735125) was cloned by PCR and RT-PCR methods, and the sequence was analysed based on the database of rubber tree Colγletotrichum gloeosporioides HBCg01 genome. The results showed that the CgAtg4 open reading frame(ORF) was 960 nt, cod-ing319aminoacidresiduewithahighlyconservedproteasedomainofPeptidase C54with213aa.Theaminoacidsequence homology of CgA tg4 with other Atg4 genes from 19 kinds of fungals was 38% to 87%. It is predictal that CgA tg4 gene is in-volved in the process of sporulation, conidiophore germation and formation of appressorium turgor pressure of rubber tree Colγletotrichum gloeosporioides.
