越南伯克霍尔德氏菌B418杀线虫活性产物的稳定性研究

         

摘要

In order to study the separation and purification of Burkholderia vietnamiensis B418 Metabolites with nematicide ability, the thermal stability, pH stability and organic solvents stability of B418 fermentation supernatant was analyzed by the method of root-knot nematode 2j larvae bioassay. The results showed that the maximum processing temperature of the active material should not exceed 60℃. pH was stable in the range from 7.0 to 10.0. When pH is 7.0, it has the highest mortality rate of 76.63% in 8 hours after treatment. The treatment of ethyl acetate, petroleum ether, acetone and n-butanol on B418 fermentation supernatant decreased the nematicidal activity. Besides, Chloroform did not affect nematicidal activity of B418 Meloidogyne spp. while the nematode mortality rate remained stable at 74%.%研究越南伯克霍尔德氏菌(Burkholderia vietnamiensis)B418产生的杀线虫活性代谢产物,采用根结线虫(Meloidogyne spp.)2龄幼虫生物测定法对B418发酵液上清液的热稳定性、酸碱稳定性及有机溶剂的稳定性进行了研究。结果表明,B418代谢产物的处理温度最高不应超过60℃;pH稳定范围为7.0~10.0,其中pH为7.0时B418代谢产物活性最高,B418代谢产物处理后8 h试虫最高校正致死率为76.63%;以乙酸乙酯、石油醚、丙酮、正丁醇均降低了B418代谢产物的杀线虫活性,氯仿不影响代谢产物活性,氯仿处理后的代谢产物线虫死亡率稳定在74%以上。
