孑遗植物双蕊兰菌根真菌培养条件的优化

     

摘要

The Orchid relict plant Diplandrorchis s inica fungi were used as materials.By separation,purification and cultured,the D.sinica mycorrhizal fungi culture system which based on the principle of orthogonal design was established,using orthogonal experimental method to set culture conditions that include temperature,carbon source,nitrogen source and medium pH.The results showed that the white or pale yellow colonies of D.sinica fungi showed the uniform radial outward growth,growth surface of sloppy or rich aerial hyphae with transparent droplets.Through measurement and analysis of colony growth rate,best level of four factors were as following:The carbon source was concentration of 2% galactose;The nitrogen source was concentration of 0.5% ammonium nitrate;pH was 6;The temperature was 25 ℃.Colony growth curve showed downwardtrend,with the increase in the number of days,and finally tends to a constant value.%以兰科孑遗植物双蕊兰(Diplandrorchis sinica)菌根为材料,通过分离、纯化、培养其菌根真菌,根据正交试验设计原理,利用正交试验方法筛选温度、碳源、氯源和培养基pH等培养条件建立双蕊兰菌根真菌培养体系.结果表明,双蕊兰菌根真菌菌落白色或浅黄色,呈均匀辐射状向外生长;生长面有稀松或较丰富的气生菌丝,带有透明液滴.通过对菌落生长速率的测定与分析,4种因素的最佳综合水平为:碳源为半乳糖,浓度为2%;氮源为硝酸铵,浓度为0.5%;pH为6;温度为25℃.菌落生长速率随着天数的递增而以较快的速度下降,第7天之后趋于稳定.
