桥本氏甲状腺炎背景下甲状腺癌中的差异表达蛋白分析

     

摘要

Objective: Objective:To analyze the high expression differential protein in Hashioto’ s thyroiditis complicated with thyroid cancer and explore the pathogenesis and provide basis for searching specific tumor marker. Method:Applying vitro labeling technique ( method of comparative pro-teomics) , protein extracts on Hashimoto's thyroiditis background patients with papillary thyroid carcinoma and follicular thyroid adenoma and Hashimoto's thyroiditis gland tissue was analyzed by iTRAQ. And the normal thyroid tissue were selected as the control group. The differential expression protein in four tissues were to be seeked. And the differential apparent protein were done with data retrieval. The biological characteristics of the differential apparent protein were identified and verified. Result:Through the analysis of four groups of protein extracts on Hashimoto's thyroiditis background of thyroid cancer, thyroid adenoma, Hashimoto's thyroiditis tis-sues and normal thyroid tissue comparison, and the protein extracts of thyroid cancer tissue obtained by the comparative analysis were coincided and analyzed. This paper achieved the differential protein and performs the superposition analysis, and finally got 8 kinds of differential expression protein with relatively high differ-ence:K6PL HUMAN, NUCB2 HUMAN, CNBP HUMAN, UB2V2 HUMAN, RAB8A HUMAN, APOA4 HUMAN, RPL13 HUMAN and TES HUMAN. Through verifying the Western Blotting of iTRAQ result, it a-chieved the verification results of the above 8 kinds of proteins. Through the comparison, it turned out that RPL13 showed the most obvious expression difference among four tissues. Conclusion:Through the compari-son on the protein, it showed that the protein with 8 kinds of expressions had relatively high difference during the occurrence of Hashioto’ s thyroiditis complicated with papillary carcinoma. However, the ribosomal protein 13 ( RPL13) showed the most obvious expression in Hashioto’ s thyroiditis complicated with papillary thyroid carcinoma, which may be the protein greatly related to the occurrence and development of Hashioto’s thyroidi-tis complicated with thyroid cancer.%目的::对桥本氏甲状腺炎背景下甲状腺癌中的高表达差异蛋白进行分析,探索桥本氏甲状腺炎背景下甲状腺癌的发病机理、为寻找特异性肿瘤标志物提供有力依据。方法:采用多肽体外标记技术(即比较蛋白质组学的方法),对桥本氏甲状腺炎背景下甲状腺乳头状癌患者的癌组织和甲状腺滤泡性腺瘤以及桥本氏甲状腺炎腺体组织的蛋白提取物进行iTRAQ分析,并与正常甲状腺组织作为对照组。寻找四种组织中差异表达的蛋白质,并将差异性明显的蛋白进行数据检索,明确这些差异性明显的蛋白质的生物学特性,并进行验证。结果:通过对桥本氏甲状腺炎背景下甲状腺癌组织、甲状腺腺瘤组织、桥本甲状腺炎组织、正常甲状腺组织的四组蛋白提取物的分析比较,比较所得差异蛋白质,进行重合分析,最后得出8种有较高差异的表达蛋白。分别是:K6PL HUMAN, NUCB2 HUMAN, CNBP HU-MAN,UB2V2 HUMAN,RAB8A HUMAN,APOA4 HUMAN,RPL13 HUMAN,TES HUMAN。对 iTRAQ结果进行western blotting验证,得出以上八种蛋白的验证结果,比较发现,RPL13在四种组织中表达差异最明显。结论:通过蛋白比对发现,在桥本氏甲状腺炎背景下甲状腺乳头状癌的发生过程中有8种表达的蛋白有较高差异,而核糖体蛋白13( RPL13)在桥本氏甲状腺炎背景下甲状腺乳头状癌组织中表达最明显,可能为桥本氏甲状腺炎背景下并发甲状腺癌的发生、发展相关性较大的蛋白质。
