结直肠癌MICA基因的表达与p53K-ras基因突变的关系研究

         

摘要

Objective:To investigate the relationship between the expression of MICA gene in bladder cancer and the mutation of p53 gene and K-ras gene.Methods: 66 cases of bladder cancer patients with clinical data in our hospital from March 2014 to March 2016 were analyzed.Semi quantitative PCR-SSCP technique was used to detect the expression of MDR1 gene and p53 and K-ras in cancer tissues,and analyzed the relationship between the expression of MICA gene in colorectal cancer and the mutation of p53 and K-ras genes.Results: The study found that there was no correlation between the expression of MICA and the location,age and sex of the patients with colorectal cancer(P > 0.05).In 66 patients,the mean expression of MICA in 22 lymph node metastasis patients was higher than that without lymph node metastasis,and the difference was statistically significant(P < 0.05).8 cases of K-ras and p53 gene expression of the MICA gene mutation combined with average weight is higher than non mutation,the difference was statistically significant(P < 0.05),12 cases of K-ras gene mutation MICA average expression was slightly higher than that without mutation,but the difference was not statistically significant(P > 0.05),16 cases of p53 gene mutation MICA expression was significantly better than the average mutation in the p53 gene,the difference was statistically significant(P < 0.05).Conclusion: The high expression of MICA gene has become a key indicator for the detection of bladder cancer,and the correlation between K-ras and p53 gene and its high expression.%目的:分析结直肠癌MICA基因的表达与p53、K-ras基因突变的关系.方法:回顾性分析我院2014年3月至2016年3月收治的66例结直肠癌患者的临床资料,应用半定量PCR-SSCP技术,对癌组织mdr1基因表达量与p53、K-ras进行检测,并分析结直肠癌MICA基因表达与p53、K-ras基因突变间的相互关系.结果:经研究发现,MICA表达与结直肠癌患者肿瘤部位、年龄、性别无相关性(P>0.05).于66例患者中,22例淋巴结转移者的MICA平均表达量明显高于淋巴结未转移者,差异具有统计学意义(P<0.05).8例K-ras、p53基因联合突变者MICA基因平均表达量高于非联合突变者,差异具有显著统计学意义(P<0.05),12例K-ras基因突变者MICA平均表达量稍高于未突变者,但比较差异无统计学意义(P>0.05),16例p53基因突变者MICA平均表达量显著优于p53基因未突变者,差异具有统计学意义(P<0.05).结论:MICA基因高表达已成为了结直肠癌的关键检测指标,而p53、K-ras基因与其高表达具有相关性.
